摘 要:目的探讨黄连对糖尿病合并脑缺血再灌注模型大鼠Bax蛋白和Bcl-2蛋白表达的影响及其保护糖尿病合并脑缺血再灌注大鼠的脑损伤作用机制。方法本实验将SD雄性大鼠60只随机分为4组,即假手术组和糖尿病脑缺血再灌注黄连高、中、低(100 g/kg,50 g/kg,25 g/kg)剂量组,每组15只。黄连干预组给予佐链脲菌素(STZ)注射造糖尿病模型。连续监测血糖7 d,待血糖稳定后,用线栓法造脑缺血再灌注模型。观察黄连对糖尿病大鼠脑缺血再灌注120 min后脑组织Bax蛋白和Bcl-2蛋白表达的影响。结果假手术组的Bax蛋白较其他各组低,差异有统计学意义(P<0.05),而黄连低、中、高剂量组的Bax蛋白表达呈降低趋势(P<0.05)。假手术组的Bcl-2蛋白表达亦较其他各组低,各黄连剂量组的Bcl-2蛋白表达随黄连剂量增加而增高(P<0.05),降低血糖,可明确改善神经功能缺损症状(P<0.05)。结论高剂量黄连能下调Bax蛋白、上调Bcl-2蛋白表达,降低血糖,改善神经功能缺损症状,抑制糖尿病合并脑缺血再灌注损伤后细胞凋亡,从而对糖尿病合并脑缺血再灌注损伤起到保护作用。Objective To explore the effect of coptidis rhizoma on the expression of Bax and Bcl-2 proteins and to explore the protective mechanism in diabetic rats complicated with cerebral ischemia after cerebral ischemia reperfusion. Methods Sixty SD rats were selected and randomized into four groups:sham group and high,medium,low dose groups of coptidis rhizome( 100 g/kg,50 g/kg,25 g/kg),each involved fifteen rats. The pretreatment group with STZ injection was the diabetic model,and the rats were continuously monitor for 7 days until the blood glucose was stable,and then the cerebral ischemia reperfusion model was made by suture method. The expressions of Bax and Bcl-2 proteins were detected120 min after cerebral ischemia/reperfusion treated by coptidis rhizoma. Results The expression of Bax protein in the sham group was significantly lower than that in other groups( P0. 05),the expression of Bax protein decreased significantly with the increasing dosage of coptidis rhizoma( P〈0. 05). The expression of Bcl-2 protein in the sham group was lower than that in other groups( P〈0. 05),and the expression of Bcl-2 protein increased with the increasing dosage of coptidis rhizoma( P〈0. 05). The pretreatment group with high dosage of coptidis rhizoma could improve the neurological deficit and blood sugar. Conclusions Coptidis rhizoma can up regulate the expression of Bcl-2 protein and down regulate the expression of Bax protein,meanwhile reduce the neurological function score and the levels of blood sugar. Coptidis rhizoma may protect the injury induced by diabetes mellitus complicated with cerebral ischemia/reperfusion through inhibiting neuron apoptosis.
