大建中汤介导ERK1/2信号通路调控脾阳虚胃癌动物模型MMP-9的表达  被引量:7

摘  要:目的:探讨大建中汤对SD大鼠脾阳虚胃癌模型胃癌组织中细胞外调节蛋白激酶1、2(ERK1、ERK2)、基质金属蛋白酶-明胶酶(MMP-9)mRNA的影响。方法:将健康SD大鼠随机分为正常组(A)、模型组(B)、大建中汤高、低剂量组(C、D组,给药剂量为生药10.8、5.4 g/kg)、西药对照组(E组,给药剂量为0.019g/kg)。A组予生理盐水,1次/d;B、C、D、E组予番泻叶水浸液灌胃、N-甲基-N'-硝基-N-亚硝基胍(MNNG)储存液自由饮用,并使其饮食失节、疲劳过度复制脾阳虚胃癌动物模型。5个月后,B组予生理盐水,1次/d;C、D组予大建中汤,1次/d;E组予环磷酰胺,1次/d。15d后,处死大鼠。应用逆转录聚合酶链式反应(RT-PCR)法,检测大鼠ERK1、ERK2、MMP-9mRNA的表达量。结果:模型组ERK1、ERK2、MMP-9mRNA的表达量明显高于正常组,差异有统计学意义(P<0.05);大建中汤高、低剂量组、西药组ERK1、ERK2、MMP-9mRNA的量明显低于模型组,差异有统计学意义(P<0.05)。结论:大建中汤可通过调节ERK1/2信号通路调控MMP-9的表达。Objective To study the effect of Dajianzhong decoction on ERK1, ERK2, MMP - 9mRNA expression in gastric tissue of rats with spleen - yang deficiency gastric cancer. Methods Fifty SD rats were divided five groups by random, including normol group ( A), model group ( B), Dajianzhong decoction high - dosage group ( C ), loe - dosage group (D) and western medicine group (E). The A group were given normal saline. The B, C, D, E were to establish rat model. After five month, the B group were given normal saline, the C, D group were given Dajianzhong decoction ( 10. 8, 5.4 g ~ kg - 1 ) every day, the E group were giv- en western medicine (0. 019 g ~ kg-I ) every day. After fiflen days, all rats were sacrificed to detect ERK1, ERK2, MMP -9mRNA expression . Results The ERK 1, ERK 2, MMP - 9mRNA expression of group B was roser than group A (P 〈 0. 05 ) but in group C, D, E was lower than group B ( P 〈 0. 05). Conclusion Dajianzhong decoction can reinforce the expression of ERK1, ERK2, MMP - 9mRNA of rats with spleen - yang deficiency gastric cancer.
