摘  要:以壳聚糖,三氧化硫脲和盐酸为反应原料,采用微波辐射的方法合成壳聚糖单胍盐酸盐。采用正交实验法通过研究反应温度,壳聚糖和三氧化硫脲的摩尔比以及反应时间对产率和胍取代度的影响确定了最佳合成工艺。采用红外光谱和核磁共振波谱仪对壳聚糖单胍盐的结构进行了表征。测试了产品的最低抑菌浓度。结果表明,采用微波辐射的方式合成壳聚糖单胍盐酸盐较传统的合成方法反应速度提高了6倍,相比于壳聚糖,壳聚糖单胍盐酸盐具有更为优异的抗菌活性。Chitosan guanidine hydrochloride was synthesized by reacting chitosan,thiourea trioxide and hydrochloric acid under microwave irradiation.The influence of reaction temperature,molar ratio of chitosan and thiourea trioxide and microwave heating time on yield and the degree of substitution was studied by orthogonal experimental method,and optimal conditions were identified.The representative microwave synthesized chitosan guanidine hydrochloride was characterized by FT-IR and 13CNMR,taking chitosan as a reference.The antimicrobial activities of chitosan guanidine hydrochloride were measured by means of the minimal inhibition concentration(MIC).The results show that the microwave irradiation method can increase the reaction rate by six times over the conventional method.Chitosan guanidine hydrochloride is effective of bacteriostasis.Compared with chitosan,chitosan guanidine hydrochloride had much better antibacterial activity.
