中介素对大鼠心肌梗死组织中TNF-αmRNA、IL-1βmRNA表达的影响

摘　　要：目的:探讨中介素对大鼠急性心肌梗死后心肌组织中肿瘤坏死因子-αmRNA(TNF-αm RN)、白介素-1βmRNA(IL-RNA)表达的影响。方法:采用SD大鼠急性心肌梗死模型40只,随机分为正常组、假手术组、心梗组、中介素干预组。各组通过心脏彩超测定术后28 d左室舒张末期内径及左室收缩末期内径的变化;分别用荧光定量PCR检测大鼠心肌组织中TNF-αmRNA、IL-1βmRNA表达。结果:与假手术组相比,心梗组和中介素干预组TNF-αmRNA、IL-1βmRNA表达增加(P<0.05);中介素干预组与心梗组比较,TNF-αmRNA、IL-1βmRNA表达增加(P<0.05);与假手术组相比,心梗组和中介素干预组LVEDD和LVESD均升高(P<0.01);中介素干预组与心梗组比较,LVEDD和LVESD均升高(P<0.01)。结论:中介素可减轻急性心肌梗死后心室重构,其机制可能与抑制心肌梗死组织的TNF-α、IL-1βmRNA表达有关。Objective：To investigate the effect of mediator on the expression of tumor necrosis factor-αmRNA and interleukin-1β mRNA in the myocardium after acute myocardial infarction in rats.Methods：A total of 40 SD rats with acute myocardial infarction were randomly divided into the normal group,the sham operation group,the myocardial infarction group,and the intermediary intervention group.The changes of left ventricular end diastolic diameter and left ventricular end systolic diameter at28 days after cardiac surgery were measured by echocardiography,and the expression of TNF-αmRNA and IL-1βmRNA in myocardium was detected by fluorescent quantitative PCR.Results：Compared with sham operation group,the expression of intermedin TNF-αmRNA and IL-1βmRNA in the myocardial infarction group and the intervention group were increased（P〈0.05）;TNF-αmRNA and IL-1βmRNA expression in the the AMI group increased when compared with the mRNA group（P〈0.05）;compared with the sham operation group,LVEDD and LVESD were increased in the myocardial infarction group and the intermediary intervention group（P〈0.01）;Both the LVEDD and the LVESD increased in the intermedin intervention group compared with the myocardial infarction group（P〈0.01）.Conclusion：Mediator can reduce ventricular remodeling after acute myocardial infarction,and its mechanism may be related to inhibiting the expression of TNF-and IL-1 beta mRNA in myocardial infarction tissue.
