复方益气消征方对肺癌A549细胞影响实验研究

摘　　要：观察复方益气消征方(YQXZF)对A549肺癌细胞裸鼠移植瘤生长的抑制作用和作用机制及干预体外A549细胞的侵袭力的影响。方法:(1)选择4种不同浓度的益气消征方联合5-氟尿嘧啶作用于体外培养的人肺癌细胞A-549细胞,应用Matrigel Boyden小室体外侵袭试验法观察两组药物对A-549细胞侵蚀性的影响;(2)选择5-氟尿嘧啶和益气消征方联合5-氟尿嘧啶灌胃,观察对负瘤小鼠的肿瘤瘤体体积差的影响。结果:(1)4组不同浓度的中药复方联合化疗药组穿膜细胞数均优于对照组,差异有统计学意义(P<0.05),其中0.5 mg/mL浓度的中药复方联合化疗药组穿膜细胞数最多;(2)对照组小鼠瘤体体积差为:(588.7±70.9)mm^3,益气消征方联合化疗组小鼠瘤体体积差为:(431.5±60.4)mm3(P<0.05),两组数据比较具有统计学差异(P<0.05)。结论:益气消征方联合5-氟尿嘧啶可以减弱人肺癌细胞A-549的侵蚀性,减少负瘤小鼠的瘤体体积差。Objective ： To observe the inhibitory effect of Yiqi Xiaozheng Decoction （ YQXZF ） on the growth of A549 lung cancer xenografts in nude mice and the mechanism of action and the effect of intervention on the invasiveness of A549 cells in vitro. Methods ： ①Select 4 different concentrations of Yiqi Xiaozheng Decoction combined with 5-fluorouracil elimination syndrome in vitro A-549 human lung cancer cells, using the matrigel boyden chamber in vitro invasion assay were used to observe the effect of two groups of drugs attack on A-549 cells ; ② the 5-fluorouracil and Yiqi Xiaozheng Decoction combined with 5-fluorouracil administered, observation influence of tumor volume of mice negative difference. Results ： The 4 groups of different concentrations of the compound traditional Chinese medicine combined with chemotherapy drug cells were better than the control group, the difference was statistically significant （ P〈0.05 ）, the compound of traditional Chinese medicine combined with chemotherapy group, the concentration of 0.5 mg/mL transmembrane cell number; the control group tumor volume difference ： （ 588.7 ± 70.9 ） mm3, and digestion syndrome of decoction combined with chemotherapy group tumor volume difference： （ 431.5 ±60.4 ）mm3 （ P〈0.05 ）, compared with two groups of data with statistical difference （P〈0.05）. Conclusion： Yiqi Xiaozheng Decoction combined with 5-fluorouraeil can reduce the erosion of human lung cancer cell A-549 and reduce the tumor volume of negative tumor mice.
