摘 要:目的观察补体片段C3f对人胚肺成纤维细胞(MRC-5)合成α-平滑肌肌动蛋白(α-SMA)的影响。方法将低中高3个不同质量浓度(15.6,62.5,250 ng·mL-1)人工合成的补体片段C3f(实验组)加入培养基中,以刺激MRC-5细胞,以细胞免疫组化法检测α-SMA在MRC-5细胞质中的表达情况。同时,设立对照组:10 ng·mL^(-1)转化生长因子-β_1(TGF-β_1)和空白组(浓度为0的C3f),进行对比研究。结果中剂量实验组与对照组(TGF-β_1)刺激MRC-5细胞合成α-SMA量分别为7.46±0.66,7.54±0.51,差异无统计学意义(P>0.05),说明2组的量相当,表明C3f具有刺激MRC-5合成α-SMA的功能。低中高3个不同质量浓度的细胞内α-SMA表达量分别为4.95±0.78,7.46±0.66和9.23±0.21,显示C3f的浓度与α-SMA的表达量呈正相关(P<0.05),表明C3f具有促进MRC-5表达α-SMA的功能。结论补体片段C3f能够刺激MRC-5细胞内α-SMA的合成增加。Objective To observe the effect of complement fragment C3f on synthesis of α-smooth muscle actin(α-SMA) in human embryonic lung fibroblasts(MRC-5).Methods MRC-5 was cultured with C3f of different concentrations(15.6,62.5,250 ng · mL-1) in low,moderate and high dosage experimental groups.And a positive control of10 ng·mL-1 transforming growth factor-β1(TGF-β1) and a negative control of 0 ng·mL-1 C3f were also set up for comparative study.Thenα-SMA was detected by immunohistochemistry.Results The moderate dose experimental group and positive control group stimulates the synthesis of α-MRC-5 cell SMA with no statistical difference(7.46 ± 0.66 vs 7.54 ± 0.51,P 〉 0.05).It demonstrated that C3f can stimulate the function of MRC-5 on synthesis of α-SMA.With the increase of C3f concentration,the expression of intracellular α-SMA in three doses experimental groups were increased for 4.95 ± 0.78,7.46 ± 0.66 and 9.23 ± 0.21,respectively,significant differences in three doses experimental groups(all P 〈 0.05).Moreover,the concentration of C3f was positively correlated with the expression of α-SMA(P 〈 0.05).C3f has the function of promoting the expression of α-SMA in MRC-5.Conclusion Complement fragment C3f can stimulate the synthesis of α-SMA in MRC-5 cells.
