摘 要:目的考察玉郎伞多糖(YLSPS)对小鼠产生的毒性反应及死亡分布并探讨YLSPS配伍环磷酰胺后的毒性变化规律。方法按照体重将小鼠随机分为2组:给药组(YLSPS:25 g·kg^(-1))及对照组(30 mL·kg^(-1)),每组20只,用最大耐受量(MTD),进行YLSPS单次灌喂给药毒性实验,2周后观察其死亡率。用均匀设计和固定环磷酰胺LD_(50)剂量联合YLSPS的不同剂量配比两种方法,按照体重将小鼠随机均匀分7组,每组10只,按各组要求以20 mL·kg^(-1)灌喂YLSPS、腹腔注射环磷酰胺,以小鼠死亡率为指标,进行急性毒性评价。结果 YLSPS的最大耐受量为25 g·kg^(-1)·d^(-1),而且无明显毒性反应。随着YLSPS给药剂量的增多,小鼠的死亡率呈下降趋势。固定环磷酰胺LD_(50)与YLSPS不同配比的减毒作用,小鼠死亡率在一定范围内随着YLSPS剂量的增加而降低,尤其在YLSPS-CTX为≥1∶1时,尤其显著。结论 YLSPS低毒安全,配伍后能使环磷酰胺的毒性降低。Objective To investigate the acute toxicity reaction and distribution mortality of Yulangsan polysaccharides(YLSPS) to healthy mice and to study the toxicity variation of YLSPS combined with cyclophosphamide(CTX).Methods The acute toxicity of YLSPS with a single irrigation was evaluated by used the maximum tolerate dose(MTD).The mice were randomly assigned as experimental group(25 g·kg-1 YLSPS) and control group(water 30 mL·kg-1),20 mice every group.After two weeks,the mortality rate was observed.The mice were randomly assigned as 7 experimental groups,10 mice every group.Respectively,every groups mice were given 20 mL · kg-1 by gavage YLSPS and intraperitoneal injection CTX as required.The toxicity changes with the death of mice as index in different groups of CTX combined with YLSPS was investigated through two methods with uniform design(UD) and fixed-dose LD50 of CTX compatibility with YLSPS.Results The MTD of YLSPS was 25 g·kg-1·d-1 and had no obvious toxicity.The mouse mortality was decreased with the increasing dose of YLSPS.The research of the attenuated toxic action with the fixed-dose procedure showed that toxicity in this compatibility of YLSPS and CTX was decreased with the increasing dose of YLSPS in a certain range,especially the dose of YLSPS was more than or equal to the dose of CTX.Conclusion YLSPS is low toxic and safety,the toxicity of CTX combined with YLSPS is reduced.
