摘 要:目的:观察假刺藤醇提物对弗氏完全佐剂(CFA)诱导的佐剂性关节炎(AA)大鼠踝关节及脾脏组织病理学的影响。方法:采用CFA诱导大鼠AA模型,给予不同剂量假刺藤醇提物干预,测定关节肿胀个数和全身炎症状态评分以及足爪肿胀度,并观察各组大鼠踝关节滑膜、软骨、关节腔、周围软组织炎症反应及脾脏等组织病理改变情况,进行综合评分。结果:与模型组比较,假刺藤醇提物能显著减少AA大鼠关节肿胀个数,降低AA大鼠全身炎症评分,显著降低AA大鼠致炎侧和继发侧足肿胀度(P<0.05或P<0.01);踝关节组织病理结果显示,假刺藤醇提物能显著抑制炎症细胞的浸润和滑膜增生以及血管翳的形成,减轻软骨及骨质损伤程度(P<0.05或P<0.01);降低AA大鼠脾脏病理组织动脉周围淋巴鞘(PALS)的细胞密度、红髓及边缘区评分(P<0.05或P<0.01)。结论:假刺藤醇提物可显著改善AA模型大鼠炎症状态、踝关节及脾脏组织病变。Objective: To observe the effect of Jiaciteng [Embelia scandens (Lour.) Mez] on histopathology of ankle and spleen tissues in rats with adjuvant arthritis (AA) which are induced by Freund's c, omplete adjuvant (CFA). Methods: The rats were immunized with the CFA. After given different doses of Embelia scandens ethanol extract, scores of joint swelling number and systemic inflammatory, state and paw swelling determiuation were observed. The ankle tissue was examined under microscope to evaluate the pathological changes of synoviurn, cartilage and articular cavity as well as soft tissue inflammation. Results: Compared with the model group, Embelia scandens can significantly reduced the number of joint swelling in AA model rats and the systemic inflammation score of the model animals, and significantly reduced the degree of inflammatory side and secondary side swelling of the model animals (P〈0.05 or 0.01). The anatomical histopathological results showed that, Embelia scandens can significantly inhibit the infiltration of inflammatory cells and synovial hypevplasia and the formation of pannus, reduce the degree of cartilage and bone damage (P〈0.05 or 0.01), significantly reduce the spleen of the spleen of rats with histopathological tissue lymph node (PALS) cell density, district score (P〈0.05 or 0.01). Conclusion: Jiaciteng can significantly improve the AA model of rat ankle joint inflammation and tissue lesions.
