调脂Ⅰ号方对动脉粥样硬化大白兔血清炎症因子及腹主动脉LDL-R、VCAM-1mRNA表达的影响  被引量:8

摘  要:目的探讨调脂Ⅰ号方抗动脉粥样硬化的可能作用机制。方法将48只雄性新西兰大白兔随机分为假手术组8只及中药高剂量组、中药低剂量组、阳性对照组、模型组各10只。除假手术组外,其余各组均饲以高脂饲料建立动脉粥样硬化模型。假手术组和模型组以10 ml/(kg·d)生理盐水灌胃。阳性对照组以30 mg/(kg·d)阿托伐他汀钙灌胃。中药高、低剂量组给予调脂Ⅰ号方12.10、3.02 g/(kg·d)灌胃。3个月后测定各组大鼠血清总胆固醇(TC)、甘油三脂(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、超氧化歧化酶(SOD)和丙二醛(MDA)含量及血清中肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)、细胞间黏附分子1(ICAM-1)、血管细胞黏附分子1(VCAM-1)的含量。RT-PCR法检测腹主动脉低密度脂蛋白胆固醇受体(LDL-R)、VCAM-1mRNA表达。结果与模型组比较,中药高、低剂量组TC、TG、LDL-C、MDA显著下降,SOD水平显著升高,中药高剂量组TNF-α、IL-6、ICAM-1、VCAM-1含量及VCAM-1mRNA表达显著降低,LDL-R mRNA表达显著升高(P<0.05或P<0.01)。与阳性对照组比较,中药高剂量组SOD含量显著升高,TNF-α水平显著降低(P<0.05)。中药高、低剂量组各指标比较差异均无统计学意义(P>0.05)。结论调脂Ⅰ号方可改善血脂、降低血清炎性因子,上调LDL-R mRNA,下调VCAM-1 mRNA,可能是其抗动脉粥样硬化的机制之一。Objective To explore the possible mechanism of Tiaozhi Fang Ⅰ against atherosclerosis. Methods Forty-eight male New Zealand white rabbits were randomized into sham operation group( n = 8),the high dose herb group( n = 10),low dose herb group( n = 10),positive control group( n = 10) and model group( n = 10). Rabbits were made atherosclerosis models by high-fat diet except for the sham operation group. The sham operation group and the model group were given 10 ml/( kg·d) normal saline by gavage. The positive control group was given 30 mg/( kg·d) atorvastatin calcium by gavage. The high and low dose herb groups were given Tiaozhi Fang Ⅰ 12. 10 and 3. 02 g/( kg·d) by gavage. After 3 months,the levels of rabbits' serum total cholesterol( TC),triglyceride( TG),low density lipoprotein cholesterol( LDL-C),high density lipoprotein cholesterol( HDL-C),superoxide dismutase( SOD),malonaldehyde( MDA),serum tumor necrosis factorα( TNF-α),interleukin 6( IL-6),intercellular adhesion molecule 1( ICAM-1) and vascular cell adhesion molecule 1( VCAM-1) in each group were determined. The expressions of aorta abdominalis low density lipoprotein cholesterol receptor( LDL-R) and VCAM-1 mRNA were detected by reverse transcription-polymerase chain reaction( RT-PCR) method. Results Compared with the model group,TC,TG,LDL-C and MDA in the high and low dose herb groups decreased; SOD level increased. The levels of TNF-α,IL-6,ICAM-1 and VCAM-1 and VCAM-1 mRNA expression in the high dose herb group decreased; LDLR mRNA expression increased( P〈0. 05 or P〈0. 01). Compared with the positive control group,SOD level in the high dose herb group increased; TNF-α level decreased( P〉0. 05). The difference of indexes between high and low dose herb groups was not significant( P〈0. 05). Conclusion Tiaozhi Fang Ⅰ might improve blood lipid,reduce serum inflammatory factor,up-regulate LDL-R mRNA and down-regulate VCAM-1 mRNA,which might be one o
