摘  要:为了探讨盐皮质激素受体基因(NR3C2基因)作为半番鸭异常行为研究候选基因的可能性,试验以半番鸭为材料,设计9对引物对NR3C2基因遗传多态性进行研究,其PCR产物经测序后,利用生物信息学软件分析SNPs位点突变前后NR3C2基因及其蛋白结构的变化。结果表明:在扩增的NR3C2基因中筛选出7个SNPs,即Intron2-G13 928T、Intron2-A13 972G、Intron2-T14 336C、Intron4-A34 930G、Intron4-T35 157G、Exon8-C198 088T和Exon9-G199 134C,其中Exon8-C198 088T为同义突变,Exon9-G199 134C为错义突变,导致编码的亮氨酸(Leu)突变为缬氨酸(Val)。说明NR3C2基因在半番鸭中存在突变位点,可能与半番鸭异常行为有关。To study the possibility of mineralocorticoid receptor gene (NR3 C2 gene)as a candidate gene for abnormal behavior study on Mule duck. Mule duck was chosen as experimental material in this experiment, and 9 pairs of primers were designed to research genetic polymorphism of NR3C2 gene. The PCR products were sequenced,and bioinformatics software was used to analyze the changes of NR3C2 gene and its protein structure between pre - and post - mutation of SNP sites. The results showed that 7 SNPs were obtained after the screening of the amplified NR3C2 gene including Intron2 - G13 928T, Intron2 - A13 972G, Intron2 - T14 336C, Introrsl - A34 930G, Intren4 - T35 157G, Exon8 - C198 088T and Exon9 - G199 134C. Among them, Exon8 - C198 088T belonged to synonymous mutation, and Exon9 - G199 134C belonged to missense mutation which resulted in the conversion of encoded leucine(Leu) to valine(Val). The results suggested that a mutation site existed in NR3C2 gene in Mule duck ,which could be in relation to the abnormal behavior of Mule duck.
