摘  要:本文以蚕豆(Vicia faba)为材料,采用药理学方法和分光光度法,测定其气孔开度和叶片内硫化氢(hydrogen sulfide,H_2S)含量以及L-/D-半胱氨酸脱巯基酶活性(L-/D-cysteine desulfhydrase,L-/DCDes)的变化,进一步确定H_2S在聚乙二醇(PEG6000)调控气孔运动信号转导中的作用.结果显示:光下H_2S清除剂次牛磺酸(hypotaurine,HT)、H_2S合成酶抑制剂羧甲氧基胺半盐酸盐(aminooxy acetic acid,AOA)和羟胺(hydroxylamine,NH2OH)以及H_2S合成酶L-/D-半胱氨酸脱巯基酶(L-/D-cysteine desulfhydrase,L-/D-CDes)分解产物丙酮酸钾(potasium pyruvate,C3H3KO3)和氨水(ammonia,NH3)均显著抑制20%PEG6000胁迫诱导的蚕豆气孔关闭;20%PEG6000能显著提高叶片的H_2S含量及L-/D-CDes活性,且HT、AOA、NH2OH和C3H3KO3+NH3均能显著逆转PEG6000所引起的叶片H_2S水平和L-/DCDes活性的升高.结果表明PEG6000胁迫可通过促进叶片内L-/D-CDes活性提高,引起H_2S生成,进而诱导蚕豆气孔关闭.The article studied the effects of H2S on stomatal closure induced by PEG6000 stress in Vicia faba by pharmacological treatments combined with spectrophotography to determine the stomatal aperture and the content of H2S in the leaves of Vicia faba. The results showed that 20 % PEG6000 significantly induced stomatal closure,and H2S scavenger HT,H2S synthesis inhibitors AOA and NH2OH and the products of L-/D-cysteine desulfhydrase( L-/D-CDes) N3H3KO3+ NH3 all inhibited PEG6000-induced stomatal closure. 20 % PEG6000 could significantly increase the H2S levels and L-/D-CDes activity in leaves of Vicia faba,and HT,AOA,NH2OH and C3H3KO3+ NH3 all could reverse the increase induced by PEG6000. These results suggested that PEG6000 induced stomatal closure by increasing H2S levels of leaves in Vicia faba.
