摘 要:为研究DF-1细胞的泛素-蛋白酶体系统(UPS)是否参与新城疫病毒(NDV)在细胞中复制的过程。使用Western-blot检测感染NDV Herts/33毒株的细胞样品以判断细胞内泛素化蛋白水平的变化,并检测NDV感染对细胞26S蛋白酶体水解活性的影响。检测泛素-蛋白酶体系统抑制剂对NDV感染后细胞上清中病毒滴度的影响,以及抑制剂MG-132对NDV在细胞中生长状况的影响。结果显示,虽然感染NDV后DF-1细胞内泛素化蛋白水平并未发生明显变化,但26S蛋白酶体的活性显著升高;使用泛素-蛋白酶体系统抑制剂可以抑制NDV在DF-1细胞中的复制。本研究结果证明DF-1细胞内泛素-蛋白酶体系统参与了NDV在DF-1细胞中的复制过程。In order to investigate whether the ubiquitin-proteasome system(UPS)of DF-1 cells was involved in the replication of Newcastle disease virus(NDV)in DF1 cells,Herts/33 strain was used to infect DF-1 cells,and the cell samples were subjected to Western-blot experiments and the three proteolytic activity changes of the 26 S proteasomes were detected.At the same time,four kinds of ubiquitinproteasome inhibitors were used after the cells were infected with NDV and the virus TCID50 values were determined.The effects of inhibitors treatment and DMSO treatment on NDV replication were compared.As well as the growth curve of NDV with the treatment of MG-132 or DMSO.The results showed that the levels of ubiquitinated proteins in DF-1 cells did not change significantly after NDV infection,but the proteolytic activity of 26 S proteasomes was significantly increased.After using ubiquitin-proteasome inhibitors the proliferation of NDV was significantly inhibited,suggesting that the ubiquitin-proteasome system of DF-1 cells was involved in the replication of NDV in DF-1 cells.
