西格列汀对2型糖尿病大鼠Nrf2表达调控及胰岛β细胞功能的影响

摘　　要：目的研究西格列汀对2型糖尿病大鼠Nrf2表达调控及胰岛β细胞功能的影响。方法雄性Wistar大鼠80只适应性饲养1 w后,采用随机数字表选取20只胰岛β细胞功能指数作为正常对照组,其余60只构建2型糖尿病模型,造模成功后根据随机数字表分为模型组和治疗组,每组20只。模型组灌胃0.9%生理盐水,治疗组灌胃0.9%生理盐水配制的西格列汀,给药剂量为100 mg·kg^(-1)·d^(-1),灌胃周期为20 w。对各组大鼠胰岛β细胞功能,Nrf2表达水平进行分析。结果干预20 w后,治疗组血糖水平显著低于模型组(P<0.05),血清胰岛素水平显著高于模型组(P<0.05)。治疗组(HBCI)、胰岛β细胞分泌功能指数(FBCI)显著均高于模型组(P<0.05)。治疗组大鼠血清和胰腺丙二醛(MDA)、肿瘤坏死因子(TNF)-α显著低于模型组(P<0.05)。治疗组大鼠血清和胰腺总超氧化物歧化酶(T-SOD)均显著高于模型组(P<0.05)。治疗组大鼠Nrf2表达水平显著高于模型组(P<0.05)。结论西格列汀可显著上调2型糖尿病大鼠Nrf2表达水平,改善其胰岛β细胞功能,对2型糖尿病具有确切的临床疗效。Objective To study the effect of sitagliptin on pancreatic beta cell function and the expression of Nrf2 in type 2 diabetic rats.Methods 80 male Wistar rats were fed for 1 week,as 20 rats were selected as normal control group according to the random number table,then construct the model of type 2 diabetes mellitus. The remaining 60 rats were randomly divided into model and treatment groups,20 rats in each group. After the success of modeling,the model group was orally administered with 0.9% saline,and the treatment group was given sitagliptin dissolved with 0.9% saline. Results After 20 weeks of treatment,the blood glucose level of treatment group was significantly lower than that of model group（ P 0.05）,serum insulin level was significantly higher than that of model group（ P〈0. 05）. The levels of HBCI,FBCI in treatment group were significantly higher than those in model group（ P〈0. 05）. The levels of serum MDA,TNF-alpha and MDA in pancreatic of treatment group were significantly lower than those in model group（ P〈0. 05）.The levels of T-SOD in serum and pancreas in treatment group were significantly higher than those in model group. The expression level of Nrf2 of treatment group was significantly higher than that of model group（ P〈0. 05）.Conclusions Sitagliptin could upregulate Nrf2 expression in type 2 diabetic rats,improve the function of islet beta cells,which has definite clinical effect on type 2 diabetes.
