气滞血瘀型输卵管炎大鼠模型的建立

摘　　要：目的采用复合刺激联合输卵管注菌的方法,制造符合中医证候特点的气滞血瘀型输卵管炎大鼠模型。方法将64只Wistar雌性大鼠随机分为对照组、输卵管炎模型组(输卵管炎组)、气滞血瘀型模型组(气滞血瘀组)、气滞血瘀型输卵管炎模型组(病证模型组)。正常对照组予以普通喂养,炎性组输卵管注菌造模,气滞血瘀组采用复合刺激干预处理,病证模型组采用复合刺激联合输卵管注菌进行造模,观察各组大鼠输卵管病理及相关指标变化情况。结果病证模型组出现气滞血瘀的症状,输卵管与周围组织粘连,显微镜下观察输卵管形态改变、增粗、积水,与其余各组血流变(血黏度、血浆黏度)及炎症指标(TNF-α、γ-IFN水平)比较,差异有统计学意义(P<0.05),与正常对照组受孕率比较,病证模型组受孕显著降低。结论复合刺激联合输卵管注菌建立的气滞血瘀型输卵管炎模型,符合人类自然发生的输卵管炎病理特点和中医气滞血瘀证候特点,是较为理想的气滞血瘀型输卵管炎模型。Objective To establish the rat model of salpingitis of qi and blood stagnation pattern/syndrome in compliance with TCM syndrome characteristics,using the compound stimulation and bacteria inoculation. Methods A total of 64 Wistar female rats were randomized into a normal control group,a salpingitis model group,a qi and blood stagnation model group and a salpingitis of qi and blood stagnation model group（syndrome model group）. In the control group,the rats were fed regularly. In the salpingitis model group,the bacteria inoculation was used for modeling. In the qi and blood stagnation model group,the intervention with compound stimulation was adopted. In the syndrome model group,the compound stimulation and bacteria inoculation were used in combination for modeling. The pathological changes of fallopian tube and relevant indexes were observed in the rats of each group. Results In the syndrome model group,there were the symptoms of qi and blood stagnation and the adhesion of fallopian with the peripheral tissues. Under microscopic observation,there were the morphological changes of fallopian tube such as the thickened tube and hydrops. The differences were significant in hemorheological indexes（blood viscosity,plasma viscosity） and inflammatory indexes（serum TNF-α,IFN-γ） as compared with the other groups（P 0. 05）. Compared with the normal control group,the conception rate in the syndrome model group was reduced significantly.Conclusion The rat model of salpingitis of qi and blood stagnation pattern/syndrome,using the compound stimulation and bacteria inoculation is in compliance with the pathological characteristics of salpingitis in human beings as well as with TCM syndrome characteristics. It is the satisfactory salpingitis model of qi and blood stagnation pattern/syndrome.
