摘 要:目的探讨不同剂量健心颗粒对慢性心衰大鼠钙调神经磷酸酶(Ca N)、尾加压素Ⅱ(UⅡ)表达的影响。方法将64只18周龄的雄性自发性高血压大鼠(SHR)随机分为阴性对照组,培哚普利组,健心颗粒等、高剂量组,每组16只。阴性对照组予生理盐水,培哚普利组给予培哚普利0.36mg/(kg·d),健心颗粒等、高剂量组大鼠分别给予健心颗粒3.2 g/(kg·d)、6.1 g/(kg·d),按1 ml/100 g灌胃给药,2次/d。给药后第21天、35天各组分别处死8只大鼠,采用蛋白质印迹法(Western blotting)检测心室肌组织Ca N、UⅡ蛋白表达。结果各组大鼠心室肌Ca N、UⅡ蛋白表达情况,第21天,阴性对照组比较与培哚普利组比较差异无统计学意义,健心颗粒等剂量组和健心颗粒高剂量组大鼠心室肌Ca N、UⅡ蛋白的表达与阴性对照组比较均显著下降,且健心颗粒等剂量组下降程度明显低于健心颗粒高剂量组(P<0.05)。第35天,与阴性对照组比较,其他各组大鼠心室肌Ca N、UⅡ蛋白的表达均显著下降(P<0.05),且存在培哚普利组<健心颗粒等剂量组<健心颗粒高剂量组。结论健心颗粒可以通过从蛋白水平影响UⅡ、Ca N的表达,抗心室重构作用显著。Objective To explore the impacts of jianxin granules of different doses on the expressions of calcineurin(Ca N) and urotensin Ⅱ(UⅡ) in the rats of chronic heart failure. Methods A total of 64 male rats of spontaneous hypertension,aged 18 weeks were randomized into a negative control group,a perindopril group,a jianxin granule isodose group and a jianxin granule high-dose group,16 rats in each one. In the negative control group,the physical saline,6. 1 g/(kg·d) was used. In the perindopril group,the jianxin granules isodose group and the jianxin granules high-dose group,perindopril,0. 36 mg/(kg · d),jianxin granules 3. 2 g/(kg·d) and 6. 1 g/(kg·d) were used separately. The medicines were dissolved with physical saline for the gavage administration,1 ml/100 g and twice a day. On day 21 and 35 after medication,8 rats were sacrificed in each group separately. The Western blotting method was used to determine the expressions of Ca N and UⅡ. Results On day 21,regarding the expressions of Ca N and UⅡin the ventricular muscle,compared with the negative control group,the differences were not obvious in the perindopril group. The results in the jianxin granules isodose group and the jianxins granule high-dose group were all reduced significantly. But the reducing degree in the jianxin granules isodose group was more obvious than that in the high-dose group(P〈0. 05). On day 35,compared with the negative control group,the expressions of Ca N and UⅡ were all reduced significantly in the rest groups. The results in the periondopril group was the jianxin granules isodose group the jianxins granule high-dose group(P〈0. 05). Conclusion Jianxingranules achieves the significant effects of anti-ventricular reconstruction through influencing the expressions of Ca N and UⅡ.
