摘  要:目的:观察调脉饮全方及拆方对2种实验性心律失常动物模型的作用及相关机制。方法:将动物随机分为模型组、调脉饮全方组、凉血清热药组、去凉血清热药组和阳性药组。采用生理记录系统观察调脉饮全方及拆方对氯化钙(CaCl_2)诱发大鼠室性心律失常、哇巴因诱发豚鼠室性心律失常的影响;采用Western Blot法检测豚鼠心肌组织钠,钾-腺苷三磷酸酶(Na^+K^+-ATP)、钙-腺苷三磷酸酶(Ca^(2+)-ATP)和电压门控钠通道5α(sodium voltage-gated channel alpha subunit 5,SCN5A)的蛋白表达。结果:调脉饮全方组和凉血清热药组均可延迟CaCl_2诱发大鼠的室性早搏和死亡时间,调脉饮各组方对于哇巴因诱发豚鼠室性早搏和死亡时间均有一定的延长作用,其中全方组和凉血清热药组延迟死亡时间较为显著,有统计学意义(P<0.05)。调脉饮各组豚鼠心肌Na^+K^+-ATP、Ca^(2+)-ATP的蛋白表达降低,SCN5A的表达无显著差异。结论:调脉饮全方组与凉血清热药组具有对抗实验性室性心律失常的作用。Objective : To observe the influence of Tiaomaiyin and its separate components on two kinds of experimental ventrieular arrhythmia in animals. Methods : The animals were randomly divided into five groups : model group, whole Tiaomaiyin group, Liangxue Qingre components group,whole Tiaomaiyin without Liangxue Qingre components group and positive control group. By using physiological recording system, we observed the effects of Tiaomaiyin and its separate components on ventricular arrhythmia induced by CaCl2 in rats and Ouabain in guinea pigs. By using Western Blot, we detected protein expressions of Na +K + -ATP, Ca2+ -ATP and sodium voltage -gated channel alpha subunit 5 (SCN5A) in Guinea pigs myocardial tissue. Result: The whole Tiaomaiyin group and Liangxue Qingre component both could delay ventricular premature beat and the time of death in rats induced by CaC12. Each Tiaomaiyin separate components all can extend ventricular premature beat and the time of death in guinea pigs induced by Ouabain. In all groups, the whole Tiao- maiyin and the components for Liangxue Qingre group could significantly delay time of death. Differences were statistical- ly significant( P 〈 0. 05 ). The protein expressions of Na+ K+- ATP and Ca2+ - ATP in Guinea pigs myocardial tissue all reduced in each of group. Protein expression of SCNSA was no significant difference. Conclusion:The whole Tiaomaiyin and the components for Liangxue Qingre groups had an antagonism on experimental ventricular arrhythmia.
