摘 要:目的:探讨β-细辛醚对Aβ_(1-42)诱导PC12细胞炎症因子表达的影响。方法:(1)将对数生长期PC12细胞随机分为正常对照组、Aβ_(1-42)3.3μM、Aβ_(1-42)10μM、Aβ_(1-42)30μM。采用实时无标记动态细胞分析技术(Real Time Cell Analusis,RTCA)和MTT法检测各组PC12细胞存活率;Western blotting检测PC12细胞的IL-1β和TNF-α蛋白表达的变化。(2)将对数生长期PC12细胞随机分为正常对照组、Aβ_(1-42)模型组、β-细辛醚18.5μg·mL^(-1)、β-细辛醚55.5μg·mL^(-1)、β-细辛醚166.7μg·mL^(-1)。采用Western blotting检测PC12细胞的IL-1β和TNF-α蛋白表达的变化。结果:(1)3.3μM Aβ_(1-42)能显著增加PC12细胞IL-1β和TNF-α的蛋白表达(P<0.05),对PC12细胞的存活率无显著影响(P>0.05)。(2)与正常对照组相比,Aβ_(1-42)模型组细胞的IL-1β和TNF-α的表达明显增加(P<0.01)。与Aβ_(1-42)模型组相比,β-细辛醚(55.5μg·mL^(-1)、166.7μg·m L^-1)能显著降低PC12细胞IL-1β、TNF-α的表达(P<0.01),β-细辛醚(18.5μg·mL^(-1))能降低PC12细胞IL-1β的表达(P<0.05),但对TNF-α的表达无显著影响(P>0.05)。结论:β-细辛醚可以有效地抑制Aβ_(1-42)诱导的PC12细胞分泌IL-1β、TNF-α的蛋白表达。Objective :To investigate the impact of β -asarone on inflammatory cytokines secreted by PC12 cells in- duced by Aβ1-42. Methods:( 1 )PC12 cells cultured in vivo were divided into 4 groups, namely control group, low dose Aβ1-42 group (3.3 μM), middle dose Aβ1-42 group (10 μM), high dose Aβ1-42 group (30 μM). Real Time Cell Analusis (RTCA) and MTT were used to evaluate the survival rate of PC12 cells. The expressions of IL - 1β and TNF - α were examined by Western blotting. (2)PC12 cells cultured in vivo were divided into 5 groups, namely control group, model group, low dose β- asarone group (18. 5 μg/mL), middle dose β -asarone group(55.5 μg/mL), high dose β - asarone group ( 166. 7 μg/mL). The expressions of IL - 1β and TNF -α were examined by Western blotting. Results : (1) The Aβ1-42 at the dose of 3.3 μM could up regulate the expressions of IL- 1βand TNF- α( P 〈 0.05 )and had no significant effect on the survival rate of PC12 cells ( P 〉 0.05 ). ( 2 ) The results showed that the expressions of IL - 1β and TNF - α were up regulated in model group compared with those of the control group(P 〈0. 01 ). The β - asarone at the dose of 55.5 μg/mL and 166. 7 μg/mL could slow down the expressions of IL - 1β and TNF - α(P 〈0. 01 ). The 13 - asarone at the dose of 18.5 μg/mL could slow down the expression of IL - 1β( P 〈 0. 05 ) and had no significant effect on the expression of TNF - α ( P 〉 0. 05 ). Conculusion : The β - asarone could significantly inhibit the expressions of of IL - 1β and TNF -α in PC12 cells induced by Aβ1-42.
