摘　　要：目的:研究中药漏芦对胃癌细胞中促癌非编码小RNA表达、以及胃癌细胞恶性行为的影响。方法:制备漏芦乙醇提取物,以不同浓度(50~150μg/m L)作用于体外培养的人胃癌细胞AGS,采用实时荧光定量PCR法检测细胞中非编码小RNA的表达水平,采用软琼脂集落形成法检测锚定非依赖性生长能力,采用划痕法检测细胞迁移能力,采用Matrigel穿膜法检测细胞侵袭能力。结果:漏芦乙醇提取物显著下调了胃癌细胞中促癌非编码小RNA mir-191-5p、miR-125b、piRNA651和piRNA932的表达水平,有力抑制了胃癌细胞的锚定非依赖性生长能力、迁移能力和侵袭能力。结论:中药漏芦能够通过下调促癌非编码小RNA的表达抑制胃癌细胞的恶性行为,在胃癌的治疗上具有良好的应用价值。Objective： To investigate the influence of Rhaponticum uniflorum on the non - coding small RNA expres- sion and the malignant phenotype of gastric cancers. Method： AGS human gastric cancer cells were cultured and treated with different concentrations （50 -150 μg/mL） of Rhapontieum uniflorum ethanol extract. The expression of non- cod- ing small RNAs was detected by Realtime PCR. The anchorage - independent growth ability was measured by soft agar colony formation experiment. The cell migration was determined by scratch wound assay, and cell invasion was evaluated by Matrigel assay. Results ： The Rhaponticum uniflorum alcohol extract significantly down - regulated the expression of the non- coding small RNA mir- 191 -5p, miR- 125b, piRNA651 and piRNA932 in AGS ceils. At the functional level, proliferation, anchorage - independent growth and invasion of AGS cells were strongly inhibited by the extract treatment. Conclusion： The Chinese herbal Rhaponticum uniflorum inhibits the malignant phenotype of gastric cancer cells by down - regulating the oncogenie non - coding small RNAs, and Rhaponticum uniflorum could be valuable for the treatment of gastric cancer.
