摘  要:目的:探讨胶质纤维酸性蛋白(GFAP)、离子钙接头蛋白(Iba-1)在阿尔茨海默病(Alzheimer's disease,AD)发病中的作用,以及心俞、肾俞麦粒灸法对其的影响。方法:鉴定APP/PS1双转基因AD传代小鼠的基因表型,选取1.5月龄雌性转基因阳性Tg6799小鼠随机分为模型组9只和治疗组8只,同龄、同背景的C57BL/6J野生型雌性小鼠9只为正常对照组。治疗组取双侧心俞、肾俞行麦粒灸治疗,每日1次,10次为1个疗程,共治疗9个疗程,每个疗程之间间隔2 d。应用免疫组化法检测小鼠额叶皮层及海马区GFAP、Iba-1的表达水平。结果:与正常组相比,模型组额叶皮层区和海马区GFAP、Iba-1的表达增强(P<0.01);经麦粒灸法治疗后,治疗组小鼠大脑额叶皮层和海马区GFAP、Iba-1表达较模型组减少(P<0.05)。结论:心俞、肾俞麦粒灸疗法可减少GFAP、Iba-1的表达,抑制AD病理过程中胶质细胞的过度激活、增生,抑制神经炎症反应。Objective : To observe the effect of grain - sized moxibustion intervention on expresion of glial fibrillary a- cidic protein and ionized calcium - binding adaptor molecule 1 in the area of cortex and hippocampus in Alzheimer' s dis- ease (AD) rats so as to explore its mechanisms underlying improvement of AD in clinic. Methods: The genotyping of transgenic mice was detected by PCR. The 1.5 months transgenic (Tg) 6799 mice were randomly divided into model group( n = 9) and treatment group (n = 8). Normal group adopted the same age and background C57BL/6J female mice (n = 9 ). Moxibustion ( ignited grain - sized moxa cone) was applied to bilateral "Xinshu" ( BL15 ) and "Shenshu" (BL23) once a day for nine courses( 10 days constitute a therapeutic course) only in treatment group. GFAP and Iba- 1 (positive area, integrate optical density) in the area of cortex and hippocampus were detected by immunohistochemical- ly method. Results : In comparison with the normal group, the expression levels of GFAP and Iba - 1 in the area of cortex and hippocampus were upregulated significantly in the model group (P 〈 0. O1 ). After 9 sessions of grain - sized moxi- bustion intervention, the expression levels of GFAP and Iba- 1 were down- regulated significantly(P 〈 O. 05 ). Conclu- sion: Grain- sized moxibustion at "Xinshu" (BL15) and "Shenshu" (BL23) can reduce the expressions of GFAP and Iba - 1 in the area of cortex and hippocampus, suggesting an improvement of AD after moxibustion intervention by restrai- ning the inflammatory reaction.
