补阳还五汤对脑出血急性期脑水肿SD大鼠动物模型的作用及意义  被引量:8

摘  要:目的:探讨不同剂量补阳还五汤对脑出血急性期脑水肿动物模型的作用,为临床治疗的转化提供思路。方法:SD大鼠随机分组:A组(空白对照组)、B组(低剂量模型组)、C组(中剂量模型组)、D组(高剂量模型组)、E组(假手术组);B、C、D和E组建立脑出血动物模型,Longa/Bederson神经功能缺损评分;3 d开始灌胃,B、C、D组补阳还五汤不同剂量,E组生理盐水,A组不予处置,14 d结束;处死测脑组织含水量;胶质细胞原代培养及鉴定;FCM和TUNEL测胶质细胞凋亡;CCK-8和EDU测胶质细胞增殖;实时定量聚合酶链反应(Realtime PCR)和蛋白印记(western blot)测胶质细胞中基质金属蛋白酶-9(MMP-9)和水孔通道蛋白4(AQP4)mRNA和蛋白表达;划痕实验和侵袭小室(Transwell)测胶质细胞迁移及侵袭能力。结果:各组比较:药物干预后mRNA和蛋白,D组最显著(P<0.05);胶质细胞凋亡,E组最多(38.35±7.02),D组变化最显著(10.19±1.33);胶质细胞增殖E组最少(3.02±1.01),D组变化最显著(18.33±4.21);胶质细胞迁移与侵袭,D组最显著(P<0.05)。结论:脑出血SD大鼠动物模型急性期脑水肿用高剂量补阳还五汤干预后胶质细胞MMP-9和AQP4均显著下调及胶质细胞活性均显著改善。Objective: To observe the effect of different doses of Buyang Huanwu Decoction on animal model of brain edema in the acute stage of cerebral hemorrhage. Method: SD rats were randomly divided into two groups: Group A (blank control), Group B (low dose model group) , Group C (middle dose model group) , Group D (high dose model group), Group E (sham operation group). Group B, C, D and E established animal model of cerebral hemorrhage. We used the Longa/bederson neural function defect score. 3 d later,we started to fill the stomach. B, C, D groups were giv- en Buyang Huanwu Decoction at different doses. Group E was given the saline. Group A refused disposal. The adminis- tration was finished 14 days later. And then we measured the water content of brain tissue and carrie on the glial cells in primary culture and identification. FCM and TUNEL tested glial cell apoptosis. CCK -8 and EDU measured glial cell proliferation. Real- time quantitative polymerase chain reaction (Reahime PCR) and Western blot (Western blot) measured glial cell matrix metalloproteinase 9 ( MMP - 9) and aquaporin channel protein 4 (AQP4) mRNA and protein expression. Scratch assay and transwell measured glial cell migration and invasion. Result : Comparsion among groups : Af- ter drug intervention, mRNA and protein in Group D was the most significant (P 〈 0. 05 ). Apoptosis of glial cells in Group E was the most (38.35 ±7.02) and the change of Group D was the most significant ( 10.19 ± 1.33 ). Glial cell proliferation in group E was the least ( 3.02 ± 1.01 ) and the change of Group D was the most significant ( 18.33± 4. 21 ). Glial cell migration and invasion of Group D was the most significant ( P 〈 0. 05 ). Conclusion : Cerebral hemor- rhage animal model of brain edema in the acute stage with high dose of Buyang Huanwu Decoction stem prognosis of glial cell MMP- 9 and AQP4 were significantly down- regulated and glial cell activity was significantly improved.
