摘 要:目的探讨琐琐葡萄黄酮对阿尔茨海默病大鼠海马组织NF-κB/IкB-α信号通路的影响及可能的作用机制。方法SD大鼠分为对照组、多奈哌齐组、AD模型组、VTF低、中、高剂量组,采用海马双侧注射Aβ_(25-35)造模,通过免疫组化、免疫印迹及RT-PCR检测NF-κBp65、IкB-α在实验大鼠海马组织内的磷酸化水平、蛋白和基因表达情况。结果 VTF对Aβ_(25-35)诱导的NF-κB激活起到了一定的抑制作用。结论 VTF通过抑制IκB-α的磷酸化实现对NF-κBp65核转位的抑制,从而保护大脑神经细胞。Objective To investigate the effects of flavonoids from Vitis vinifera L. on NF-κB/IкB-α signal pathway in hippocampus of Alzheimer's rat and its possible mechanism. Methods SD rats were divided into control group,donepezil group,model group,group with low,medium and high dose of VTF. The AD rat models were established with Aβ_(25-35) by bilateral hippocampus injection. Immunohistochemistry,Western blot and RT-PCR were used to detect the phosphorylation level,protein and gene expression of NF-κBp65 and IкB-α in rats ' hippocampus. Results VTF could inhibit the activation of NF-κB induced by Aβ_(25-35). Conclusion VTF inhibited the nuclear translocation of NF-κBp65 by inhibiting the phosphorylation of IкB-α,thereby VTF could protect brain neurons.
