摘  要:目的筛选金银花体外抑制乙酰胆碱酯酶的活性部位,并对其化学成分进行分析。方法金银花提取物经反相C_(18)填料进行固相萃取(SPE)制备不同浓度甲醇梯度洗脱部位,以体外抑制乙酰胆碱酯酶能力评价其抗阿尔兹海默症作用,采用高效液相色谱-质谱联用技术对金银花化学成分进行分析。结果金银花提取物的30%甲醇洗脱部位的乙酰胆碱酯酶酶抑制活性明显强于其他洗脱部位,并存在明显的量效关系,为其体外抑制乙酰胆碱酯酶的活性部位,经液质联用分析共鉴定出14种化合物成分。结论金银花具有体外抑制乙酰胆碱酯酶活性,其主要有效部位为30%甲醇洗脱部位,具有一定的抗阿尔兹海默症的作用。Objective Screening honeysuckle in vitro inhibition of acetylcholinesterase active site,and to analyze the chemical composition. Methods The method of honeysuckle extract by reverse phase C_(18) material subjected to solid phase extraction( SPE) was prepared with different concentrations of methanol alcohol gradient site,in vitro inhibition of acetylcholinesterase ability to evaluate the effect of anti-Alzheimer's disease,high performance liquid chromatography-mass spectrometry coupled with honeysuckle chemical composition analysis. Results Honeysuckle extract water will result in 30% methanol site acetylcholinesterase inhibitory activity was significantly stronger than other parts of the elution,and there is a clear dose-effect relationship for the honeysuckle in vitro inhibition of acetylcholinesterase active site,the LC-MS analysis identified a total of 14 phenolic acids.Conclusion Honeysuckle in vitro inhibition of acetylcholinesterase activity,its main active ingredient chlorogenic acid represented by phenolic acids,has a certain anti-Alzheimer's effect.
