摘  要:目的以桂北金槐Sophora japonica cv.jinhuai为原材料,优选出槲皮素的提取工艺,为该药材的资源利用提供依据。方法选取槲皮素含量为因变量,在单因素试验的基础上,选取超声时间,料液比,超声功率为自变量,通过响应面法优化超声波辅助提取槲皮素工艺并进行预测分析。结果采用HPLC测定槲皮素含量,流动相甲醇(A)-0.5%磷酸水(B)梯度洗脱(1~12min,10%~75%A,12~24min,75%~88%A,24~35min,88%~95%A),检测波长为360nm。最佳提取工艺条件为超声时间60min,料液比为1:10,超声功率为350W。在此条件下槲皮素的提取率预测值为9.6215%,验证值为9.97%,与预测值接近。结论优选的提取工艺稳定可行、预测性良好,提高了金槐槐米的合理应用,为槐米槲皮素的制剂开发提供参考。Objective To select the extraction process of quercetin from the Sophora japonica cv. jinhuai in northern Guangxi,and to provide reference for the development of the medicinal materials. Methods The content of quercetin was selected as the dependent variable,and on the basis of single factor test,the ultrasonic time,ultrasonic power and the ratio of material to liquid were selected as independent variables,and the ultrasonic assisted extraction of quercetin was optimized by response surface method. Results The content of quercetin was determined by HPLC,and the liquid phase methanol( A)-0. 5% phosphate water( B) Gradient elution( 1 ~ 12 min,10% ~ 75% A,12 ~ 24 min,75% ~ 88% A,24 ~ 35 min,88% ~ 95% A),and the detection wavelength is 360 nm. The optimum extraction conditions for ultrasonic power 350 W,ultrasonic time 60 min. The material liquid ratio is 1: 10. Under this condition,the prediction value of quercetin was 9. 62153%,and the verification value was 9. 97%,which was close to the predictive value. Conclusion The optimum extraction process is stable,feasible and good,and the rational utilization of Sophora japonica cv. jinhuai is improved,which provides reference for the development of Sophora japonica.
