摘 要:目的葛根素磷脂复合物的最佳工艺优化,并对其进行表征。方法以葛根素与磷脂的复合率为评价指标,采用正交实验设计考察了药物反应物的质量浓度、反应投料比和反应时间这三个因素对磷脂复合物的影响,并筛选最优制备工艺,并通过UV、IR和TLC分析技术对复合物结构进行分析。结果确定葛根素磷脂复合物的最佳制备工艺为:葛根素与磷脂的投料比为1:4(w/w),反应物质量浓度为80mg·m L^(-1),反应时间2 h,该工艺条件制备的磷脂复合物复合率为100%。结论葛根素磷脂复合物的形成受葛根素的质量浓度和投料比的影响较大,反应时间也有一定的影响,且在形成复合物的过程中并没有发生化学变化,但葛根素和磷脂间有一定的作用力,与单纯的物理混合不同。Objective This study was designed for the preparation of Puerarin phospholipid complex,seeking for the optimum preparation and characterization of it. Methods The single actor experiment and orthogonal experiment were used to design to screen the optimal reparation process,which were assessed by recombination rate. In the study,three factors were explored,which were the drug mass concentration,reaction time and reaction ratio. At the same time,UV,IR and TLC were used to analyze the structure of phospholipid complex. Results The optimal preparation conditions of puerarin phospholipid complex were as follows: the quality ratio of puerarin and phospholipid was 1: 4,the drug mass concentration was 80 mg·m L^-1,and the reaction time was 2 h. When doing as the optimal preparation conditions,the highest recombination rate was 100%. Conclusion The reaction ratio and the mass concentration of puerarin had greater impact on the formation of puerarin complex,and the reaction time also had some impact. Chemical changes did not occur in the formation process,but some intermolecular forces occurred between puerarin and the phospholipid,and the phospholipid complex was essentially different with the simple physical mixtures.
