摘 要:以蓝莓叶为原料,采用60%甲醇溶液、60%乙醇溶液、60%丙酮溶液、乙酸乙酯4种不同的提取剂提取黄酮类化合物,提取条件为1:20的料液比,75℃恒温回流提取1 h,研究不同提取剂对蓝莓叶黄酮类化合物提取率的影响,结果显示60%甲醇溶液提取黄酮类化合物的提取率最高.采用大孔吸附树脂法对蓝莓叶浓缩提取液黄酮类化合物进行分离纯化,用60%甲醇溶液、60%乙醇溶液、60%丙酮溶液、三氯甲烷与甲醇(1∶1)4种不同的洗脱剂进行洗脱,比较不同溶剂洗脱黄酮类化合物的最大吸收峰及峰面积大小,结果显示,4种不同洗脱液在200~400 nm均出现2个吸收峰,但60%乙醇溶液洗脱的峰面积最大,确定60%乙醇溶液为黄酮类化合物的最佳洗脱剂;利用红外光谱法测定不同洗脱液浓缩液,显示不同洗脱液主要成分为黄酮类化合物.The blueberry leaf was raw material, using four different extracting agents such as 60% methanol solution, 60% ethanol solution, 60% acetone and ethyl acetate solution to extract flavonoids, extraction conditions was that liquid ratiol :20, reflux extraction constanted at tempreture75 ℃ for one hour. The effect of different extraction agents for blueberry leaf flavonoids to extraction yield was researched. The results showed that extraction yield of 60% methanol solution extracted flavonoids was the highest; By macroporous adsorption resin method, concentrated blueberry leaf extract flavonoid compounds was separated and purified with four different elution such as 60% methanol solution and 60% ethanol solution, 60% acetone solution, chloroform and methanol (1:1 ) eluent, maximum ab- sorption peak and peak area size of flavonoids using different solvent elution was compared. The results showed that there were two absorption peaks in 200 -400 nm, but the elution peak area was the largest of 60% ethanol solution, therefore 60% ethanol solution was the best eluent flavonoids. By infrared spectrometry different eluent concentrates were detected, main component was flavonoids.
