摘 要:为建立兰州百合试管苗规范化生产技术体系,以兰州百合鳞片为外植体,MS为基本培养基,对外植体消毒时间、鳞片放置方式,以及初代、继一代和继二代激素α-萘乙酸(NAA)和6-苄氨基腺嘌呤(6-BA)浓度等影响不定芽产生的3个主要因素进行研究。结果表明:0.1%的氯化汞最佳灭菌时间12 min,与鳞片内侧相比,鳞片外侧小突起、不定芽、小突起+不定芽个数也均以12 min消毒处理效果最佳;与鳞片外侧和近轴面向上放置相比,鳞片内侧和近轴面向下放置更有利于芽的诱导;最佳诱导和继一代培养基均为MS+NAA 0.1 mg/L+6-BA 1.0 mg/L,诱导培养30 d左右后不定芽数达7.00个,转绿率和发芽率分别为80.83%和55.00%;继一代发芽率和生叶率最高时可达45.02%和78.00%;兰州百合继二代培养时适当降低6-BA浓度,不仅不影响组培苗正常的生长发育,反而使叶片数和不定芽数达到23.67片和5.67个,以MS+NAA 0.1 mg/L+6-BA 0.5 mg/L为适宜培养基。In order to establish the standardized production technology system of Lanzhou lily (Lilium davidii var. unicolor)test-tube plantlets,the author took the scale of Lanzhou lily as the explant,used MS as the basic culture medium,and studiedthe effects of three factors (explant sterilization time,mode of placing scales,and concentration of NAA and 6-BA)on the inducement of adventitious buds. The results showed that the optimum time of explant sterilization with 0.1% HgCl2 was 12 min,and the number of both small protuberances and adventitious buds in the outside of scale was the highest under this condition. Placing scale with downward adaxial side was more advantageous to the inducement of adventitious buds than placing scale with upward adaxial side. The optimum medium for both the inducement and the first subculture of adventitious buds was MS+NAA 0.1mg/ L+6-BA 1.0 mg/ L;after inducement culture for about 30 days,the number of adventitious buds reached 7.00,and the green-turning rate and germination rate were 80.83% and 55.00%,respectively;in the first subculture,the germination rate and leafproducingrate could reach 45.02% and 78.00%,respectively. The best medium for the second subculture of Lanzhou lily was MS+NAA 0.1 mg/ L+6-BA 0.5 mg/ L,and the number of leaves and adventitious buds reached 23.67 and 5.67 separately under thiscondition.
