摘 要:果胶甲酯酶抑制因子(PMEI)能够抑制果胶甲酯酶(PME)的脱酯化作用,从而调控果胶甲酯酶的活性。本文在桃(Prunus persica)中鉴定出30个PpPMEI基因家族成员并进行了生物信息学分析,构建进化树发现,其与拟南芥PMEI基因家族进化关系较远。对PpPMEI进行蛋白性质分析发现,大多数PpPMEI蛋白存在信号肽,等电点在4.44~10.02之间,亚细胞定位表明多定位于分泌通路。部分PpPMEI表达模式分析表明,其在花、果实、叶中有表达差异,Prupe.1G118800和Prupe.5G112600在果实发育过程中表达量下降。赤霉素(GA)、脱落酸(ABA)和水杨酸(SA)三种激素处理后,Prupe.5G048900和Prupe.5G112600被GA和ABA诱导表达,Prupe.7G193700的表达被GA抑制。本文为研究PpPMEI在调控桃的花与果实发育以及激素响应等方面提供参考。Pectin methylesterase inhibitor(PMEI) can inhibit the de-esterification of the functions of pectin methylesterase(PME), and then regulate the activity of PME. Thirty PpPMEI genes in peach(Prunus persica) were identified in this study. PpPMEI proteins are not closer to At PMEIs. Bioinformatic analysis shows that 26 PpPMEI proteins have signal peptides, the theoretical isoelectric points were between 4.44 and 10.02, and most of PpPMEI proteins were localized in secretory pathway. Using quantitative real-time PCR, we found that PpPMEI were expressed differently in flower, fruit and leaf. Furthermore, the mean expression levels of two PpPMEI genes(Prupe.1 G118800 and Prupe.5 G112600) decreased with the growth of young fruit. Prupe.5 G048900 and Prupe.5 G112600 could be induced by gibberellin(GA) and salicylic acid in peach, however, Prupe.7 G193700 was inhibited by GA. Therefore, PpPMEI genes may paly an important role in the process of growth and development in peach. These results would be useful for the study in this field.
