摘  要:目的探讨中药茯苓对正常大鼠肾脏及尿液水通道蛋白-2(AQP2)水平的影响。方法 30只健康SD大鼠随机分成对照组和茯苓组各15只,用药后观察大鼠尿量,用RT-PCR半定量法检测肾内髓质AQP2及血管加压素-2型受体(V2-R)基因mRNA水平,用ELISA法检测尿液AQP2浓度,Western blot检测肾内髓质AQP2含量。结果茯苓组大鼠:尿量较对照组显著增多,尿渗量低于对照组,尿液中AQP2水平明显降低;肾内髓质AQP2、V2-R基因mRNA水平较对照组降低(P<0.05),AQP2表达亦减少(P<0.05)。结论茯苓可以抑制正常大鼠肾内髓质AQP2mRNA水平和AQP2的表达,同时减少肾内AQP2经尿液的排出。Objective To discuss the influence of Chinese medicine poria cocos on kidney aquaporin-2(AQP2)level of normal rats.Methods In total,30 healthy SD rats were divided into two groups:the normal control groupand the poria cocos group(15 each).After administration of drugs,the urine amount was observed;RT-PCR semi-quantitative analysis was used to detect kidney inner medulla AQP2 and vasopressin-2 type(V2 receptor)mRNA level,enzyme-linked immunosorbent assay(ELISA method)for detecting AQP2 concentration in the urine,Western blot was used to detect the content of AQP2 in renal medulla.Results The urine volume of rats from the poria cocos group was significantly increased compared with the control group,and the urine infiltration was lower than the control group;The level of AQP2 in the urine were significantly reduced;AQP2 level in urine decreased evidently.RT-PCR semi-quantitative analysis showed that AQP2、V2 receptor mRNA of the poria cocos group was lower than that of the control group(P <0.05).AQP2 expression was also reduced(P <0.05).Conclusion Poria cocos can inhibit the expression of AQP2 mRNA level and AQP2 in normal rat kidney,at the same time,it reduces the excretion of AQP2 in the kidney.
