摘 要:[目的]研究不同浓度纳米碳对平陆百合鳞片分化的影响,旨在优化筛选分化培养基的同时,为纳米碳应用于植物组织培养提供参考依据。[方法]以平陆百合脱毒苗鳞片为外植体,采用正交试验设计法研究了植物激素与不同浓度纳米碳配比对诱导平陆百鳞片分化的影响。[结果]6-苄氨基腺嘌呤(6-BA)、萘乙酸(NAA)和不同浓度纳米碳配比时,鳞片分化的最佳培养基为MS+6-BA 0.5 mg·L^(-1)+NAA 0.2 mg·L^(-1)+纳米碳800 mg·L^(-1),分化率达98.09%;玉米素(ZT)、吲哚丁酸(IBA)和不同浓度纳米碳配比时,鳞片分化的最佳培养基为MS+ZT 0.8mg·L^(-1)+IBA 0.3mg·L^(-1)+纳米碳600mg·L^(-1),分化率达91.43%。[结论]培养基中添加不同浓度的纳米碳均可促进百合鳞片的分化,不同浓度纳米碳对鳞片分化的影响程度不同,其最适宜添加浓度为600~800mg·L^(-1)。[Objective]The effects of different concentrations of nano-carbon on the differentiation of Lilium davidii were studied,the aim of this study was to optimize the screening medium and to provide reference for the application of nano-carbon in plant tissue culture.[Methods]The effects of plant hormones and different concentrations of nano-carbon on the differentiation of cultivated flora were studied by orthogonal design.[Result]When 6-BA,NAA and different concentrations of nano-carbon were used,the best medium for scale differentiation was MS+6-BA 0.5 mg·L^-1+NAA 0.2 mg·L^-1+nano-carbon 800 mg·L^-1,the differentiation rate of 98.09%;When ZT,IBA and different concentrations of nano-carbon were used,the best medium for scale differentiation was MS+ZT 0.8 mg·L^-1+IBA0.3 mg·L^-1+carbon 600 mg·L^-1,the differentiation rate of 91.43%.[Conclusion]The addition of different concentrations of nano-carbon in the culture medium could promote the differentiation of lily scales.The effects of different concentrations of nano-carbon on the differentiation of scales were different,and the optimum concentration was600-800 mg·L^-1.
