摘  要:目的:观察针刀干预对帕金森病(PD)大鼠行为学和纹状体神经递质DA、NA、5-HT含量的影响。方法:SD雄性大鼠,随机抽取16只作空白组,剩余采用右侧纹状体双靶点注射法成功制备PD大鼠模型后,随机分为模型组、电针组、针刀组。电针组电针"百会""太阳",每周3次;针刀组松解C_1、C_2横突后结节,每周2次。4周观察期后,采用APO诱导的旋转实验观察大鼠行为学变化,HE染色法光镜下观察中脑黑质形态学变化,高效液相色谱-电化学法检测纹状体DA、NA、5-HT的含量。结果:空白组不转圈,模型组转圈无变化,电针组和针刀组干预后转圈数较模型组减少显著(P<0.01);模型组纹状体递质DA、NA、5-HT含量显著低于空白组(P<0.01),针刀组DA、NA、5-HT含量较模型组升高显著(P<0.01或P<0.05)。结论:针刀干预可减轻PD大鼠的行为学症状和升高纹状体神经递质DA、NA、5-HT含量,缓解帕金森病症状。Objective: To observe effect of the acupotomy intervention on behavior and striatum neurotransmitters,such as contents of DA,NA,and 5-HT in PD rats. Methods: 16 SD rats( male) were randomly selected into the blank control group; the rest rats were modeled into PD rats with the method of right striatum double target injection,the PD rats were randomly divided into the model group,the electro-acupuncture group,and the acupotomy group. The rats in the electro-acupuncture group were needling on GV20 and EX-HN5,three times a week; acupotomy was used to debonding transverse processes of C_1 and C_2,twice per week. After four-week observation period,behavior changes were assessed by rotation test triggered by APO; morphological changes of substantia nigra were observed with HE staining method under light microscope; the content of DA,NA,and 5-HT were detected with HPLC-ECD. Results: In terms of the rotation test,the rats in the blank control group did not rotate; the rotation in the model group showed no change; the rotation was significantly decreased after electro-acupuncture intervention and acupotomy intervention,when compared to the model group( P < 0. 01). In terms of the content of DA,NA,and 5-HT,they significantly became lower in the model group when compared to the blank control group( P < 0. 01); they were obviously increased after acupotomy intervention( P < 0. 01,or P < 0. 05). Conclusion: Acupotomy intervention can alleviate behavioral symptoms and increase the contents of DA,NA,and 5-HT these striatum neurotransmitters when treating PD in rats.
