摘 要:目的观察磷酸化抗原(HDMAPP)、结核杆菌耐热抗原(Mtb-HAg)和结核杆菌低分子耐热抗原Mtb-HAg(10 k D)刺激结核分枝杆菌潜伏性感染者(LTBI)和TB患者外周血T细胞亚群产生IFN-γ的反应能力方面的特点及差异。方法采集LTBI和TB患者外周血,分别用HDMAPP、Mtb-HAg和Mtb-HAg(10 k D)刺激培养14 h,再加入莫能霉素继续培养6 h,收集细胞,加入荧光标记单抗进行表面分子和胞内染色,然后在流式细胞仪上检测各实验组中CD3+αβ细胞和CD3+γδ细胞胞内IFN-γ的表达。结果各抗原刺激组T细胞亚群均能产生IFN-γ,各抗原刺激组LTBI和TB患者的外周血γδT细胞中产生IFN-γ的细胞均显著高于αβT细胞;LTBIγδT细胞中产生IFN-γ细胞均比TB患者高,上述差异均有统计学意义(P<0.05)。结论γδT细胞的IFN-γ在LTBI和TB患者的不同,有望为区分LTBI和TB患者之间的差异提供新的指标。Objective To observe the characteristics and differences of reaction ability in T cell subpopulations producing IFN-γ in latent infected people( LTBI) and TB patients,when HDMAPP,Mycobacterium tuberculosis heat-resistant antigen( Mtb-HAg) and low molecular peptide antigen of Mycobacterium tuberculosis( Mtb) were used to stimulate the peripheral blood T cells.Methods The peripheral blood cells from LTBI and TB patients were collected for the stimulation and culturing with HDMAPP and Mtb-HAg and Mtb-HAg( 10 k D) for 14 hours,and then adding Monensin to cultivate for another 6 hours. Cells were collected and stained with different fluorochrome-conjugated m Abs for staining of surface molecule and intracellular cytokines. Then the expressions of IFN-γ of CD3~+αβ cells and CD3~+γδ cells among the experimental groups were detected by flow cytometry.Results The T cell subpopulations among antigen experimental groups can all produce IFN-γ. The γδ T cells producing IFN-γ were more than αβ cells among the LTBI antigen experimental groups( P〈 0. 05). The γδ T cells producing IFN-γ in LTBI patients were more than TB patients,with all the differences above statistically significant( P〈 0. 05). Conclusion The difference of γδ T cells producing IFN-γ between in LTBI and TB patients is expected to provide new indicators for distinguishing latent infected people and TB patients.
