三种特殊染色方法在显示系统性硬化症小鼠肺组织胶原纤维的比较

摘　　要：目的比较Masson染色、Sirius Red染色、Van Gieson染色用于系统性硬化症小鼠肺组织胶原纤维的染色效果。方法 BALB/C小鼠8只皮下注射博莱霉素建立系统性硬化症小鼠模型,实验结束后取左肺制成石蜡切片进行Masson染色、Sirius Red染色、Van Gieson染色,使用图像分析软件进行胶原含量定量。结果 Masson染色、Sirius Red染色、Van Gieson染色均能清楚显示胶原纤维,3种方法测定胶原纤维含量在统计学上无差异(P>0.05);Sirius Red染色较Masson染色操作简便,较Van Gieson染色更易保存,同时在偏光显微镜下能进行胶原定型。结论 Sirius Red染色更适合用来判断系统性硬化症肺组织纤维化。Objective To compare the dyeing effect of Masson staining,Sirius Red staining and Van Gieson staining for collagen fibers in the lung tissue of systemic scleroderma mice.Methods 8 BALB/C mice were subcutaneously injected with Bleomycin to establish a systemic sclero-derma mouse model.The left lung was made into paraffin sections for Masson staining,Sirius Red staining and Van Gieson staining after the experiment and the image analysis software was used to quantitate the collagen content.Results The Masson staining, Sirius Red staining and Van Gieson staining could clearly show the collagen fibers. It had no statistically significant difference in the collagen fibers determined by the three methods（P〉0.05） .The Sirius Red stai-ning was easier to use than Masson staining,easier to preserve than Van Gieson staining,and it could be used to perform collagen stereotypes under polarized light microscopy.Conclusion The Srius Red staining is more suitable for judging systemic scleroderma lung tissue fibrosis.
