微小RNA-152介导叶酸对体外血管平滑肌细胞增殖迁移的抑制作用  被引量:4

摘  要:目的探究叶酸对(folic acid,FA)血小板源生长因子(PDGF-BB)诱导的血管平滑肌细胞(VSMCs)增殖迁移的影响及其机制。方法体外用PDGF-BB诱导大鼠血管平滑肌细胞系A7r5增殖迁移,分为对照组、PDGF组、PDGF+FA组、PDGF+FA+miRNA-152 inhibitor组、PDGF+FA+miRNA-152 NC组。采用实时定量聚合酶链反应(RT-PCR)检测各组VSMCs内miR-152的表达水平。采用CCK-8试剂盒检测各组VSMCs增殖水平。采用划痕实验检测各组VSMCs迁移水平。采用蛋白印迹法检测细胞增殖核抗原(PCNA)蛋白、基质金属蛋白酶9(MMP-9)的表达水平。结果 RT-PCR结果显示PDGF干预降低VSMCs miR-152表达水平,而PDGF+FA组VSMCs miR-152表达水平显著高于PDGF组(P<0.05)。PDGF干预VSMCs的增殖迁移能力显著增强相对于对照组(P<0.05),而PDGF+FA组的VSMCs增殖迁移能力低于PDGF组(P<0.05),PDGF+FA+miR-152 inhibitor组的VSMCs增殖迁移能力强于PDGF+FA组(P<0.05)。结论 miR-152参与介导FA抑制PDGF-BB诱导的VSMCs增殖迁移作用。[营养学报,2017,39(5):484-488]Objective To explore the effects of folic acid on vascular smooth muscle cells (VSMCs) proliferation and migration induced by platelet-derived growth factor (PDGF-BB) and the mechanism involved. Methods PDGF-BB was used to induce VSMCs A7r5 proliferation and migration, and VSMCs were treated as follows: control group, PDGF group, PDGF+folic acid group, PDGF+folic acid+miRNA-152 inhibitor group, PDGF+folic acid+miRNA-152 NC group. Real-time quantitative polymerase chain reaction (Real-time PCR) was used to detect the expression of miR-152. The proliferation of VSMCs was detected by CCK-8 kit assay. Wound scratch assay was used to detect the migration of VSMCs. Western blot was used to detect the protein expression of proliferating cell nuclear antigen (PCNA) and matrix metalloproteinase-9 (MMP-9). Results Real-time PCR results showed that PDGF reduced the expression of miR-152 in VSMCs, while its expression was significantly higher in PDGF+folic acid group (P〈0.05). VSMCs proliferation and migration ability was significantly increased in PDGF group compared with that in the control group (P〈0.05) , while lower in PDGF + folic acid group than that in PDGF group (P〈0.05) . The VSMCs proliferation and migration ability of PDGF + folic acid + miR-152 inhibitor group was stronger than that of PDGF +folic acid group (P 〈0.05). Conclusion It is concluded that miR-152 participates in folic acid mediated inhibition of proliferation and migration induced by PDGF-BB in VSMCs.
