摘  要:目的:预测和分析人颗粒溶素(Granulysin,GNLY)蛋白的结构与功能。方法:采用生物信息学的方法对GNLY进行系统分析与研究,包括GNLY的理化性质、跨膜区和引导肽、亚细胞定位、二级结构功能域、蛋白质的功能分类预测、多重序列比对与系统发育树构建、三级结构建模等。结果:GNLY蛋白分子量为15.33k D,等电点为9.35,具有引导肽(N端1-22个氨基酸序列)无跨膜区,该蛋白属于分泌性蛋白;其二级结构主要为α螺旋和无规则卷曲,包含1个Sap B结构域,三级结构显示结果与二级结构预测结果相符。蛋白质多重序列比对和聚类分析显示,人GNLY蛋白与牛、猪及艾鼬等哺乳动物的GNLY蛋白具有一定的同源性。结论:GNLY生物信息学数据的探索,可为GNLY蛋白的功能研究提供参考资料和理论依据。Objective To study the structure and biological function of the human granulysin. Method: Many bioinformatics methodswere used to analysis physical and chemical properties, hydrophobicity, transmembrane region, signal peptide, secondary structure, functionaldomain and assortment, multiple alignment, phylogenetic tree and three-dimensional structures of GNLY. Result: The results showed that GNLYwas hydrophilic protein, whose calculated molecular mass was 15.33 kD, and theoretical isoelectric point was 9.35, with one signal peptide (the N-terminal 1-22 amino acid) and without transmembrane regions. The protein secondary structure were a-helix and random coil and contains a SapBmotif. The three -demensional structure prediction showed similar results with secondary structure prediction. The multiple alignment andclustering analysis showed that GNLY is an orthologous protein belong to saposin-like protein (SAPLIP)family, and has similarity to Bos taurus,Sus scrofa and Mustela putorius furo GNLY. Conclusion: The bioinformatics analysis will be helpful for the further study of the function of GNLY.
