摘　　要：目的探讨异基因造血干细胞移植术后Th9细胞重建特点。方法选取接受造血干细胞移植治疗的患者64例为研究对象,其中全相合组31例,单倍体组33例,20名健康人作为对照组,通过FCM和ELISA法动态检测患者及对照组外周血中Th9细胞和IL-9、TGF-β、IL-4的变化特点。结果全相合组和单倍体组的Th9细胞百分比值分别在移植后90d和150d基本恢复正常(P>0.05),IL-9的动态变化趋势与Th9细胞的变化相一致,全相合组IL-9的浓度在移植后60d和90d时高于单倍体组(P<0.05);全相合组TGF-β和IL-4在移植后90d和60d逐渐恢复至正常水平,单倍体组TGF-β和IL-4在移植后150d和60d恢复至正常水平;移植后发生aGVHD患者Th9细胞及IL-9和TGF-β浓度明显低于未发生aGVHD者,差异均有统计学意义(P<0.05)。移植后IL-9的浓度与TGF-β呈正相关。结论全相合组Th9细胞和IL-9的重建早于单倍体组,aGVHD患者Th9/IL-9重建延迟,IL-9可能有助于诱导机体免疫耐受。Objective To investigate the characteristics of Th9 cell reconstitution after allogeneic hematopoietic stem cell transplantation.Methods 64 patients with hematopoietic stem cell transplantation treatment and 20 healthy controls were enrolled in this study.Peripheral blood Th9 cells,IL-9,TGF-βand IL-4 from patients and controls were dynamically detected by Flow cytometry and ELISA.Results Th9 cells recovery started from day 90 and 150 in HLA-matched group and HLA-haploidentical group respectively(P >0.05);The dynamic trend of IL-9 was consistent with the changes of Th9 cells.The concentration of IL-9 in HLA-matched group were higher than that in HLA-haploidentical group at day 60 and day 90(P <0.05).TGF-βand IL-4 gradually recovered to normal levels at day 60 and day 90 after transplantation in HLA-matched group.In HLA-haploidentical group,TGF-βand IL-4 returned to normal levels at 60 dand150 dafter transplantation;The number of Th9 cell and concentration of IL-9,TGF-βin patients with acute graft-versus-host disease(aGVHD)were significantly lower than patients without aGVHD after transplantation,which the differences were statistically significant(P <0.05).The concentration of IL-9 was positively correlated with TGF-βafber transplantation.Conclusion HLA-matched group showed earlier reconstitution of Th9 cells and IL-9 than HLA-haploidentical group;the reconstitution of Th9/IL-9 was delayed in aGVHD patients,and IL-9 might be helpful to induce immune tolerance.
