摘 要:目的:基于"肾脑相济"理论观察补肾益脑电针疗法对快速老化SAMP8小鼠皮层神经元病理形态改变,探讨补肾益脑电针疗法对阿尔茨海默病(AD)的治疗作用及机制。方法:根据"肾脑相济"理论,8月龄SAMR1和SAMP8小鼠电针"百会"、"肾俞"、"太溪"穴,连续8周后,取一半脑制作石蜡切片,甲苯胺蓝染色观察各组小鼠皮层神经元内尼氏小体结构的变化。结果:对照组SAMR1小鼠皮层神经元体积较大,形态规则,胞浆内布满尼氏体;模型组SAMP8小鼠皮层神经元体积较小,有较多细胞失去完整形态,尼氏小体颜色变浅且数目减少;电针和西药组的SAMP8小鼠皮层神经元体积变大,尼氏体较致密,无明显缺失和空泡变性。结论:依据"肾脑相济"理论的补肾益脑电针疗法可改善SAMP8小鼠脑内神经元的形态和功能状态,对AD可能有很好的治疗作用。Objective To observe the effect of electroacupuncture with Bushenyinao on pathological morphological changes in the temporal cortex neurons of SAMP8 mice,and to explore the function and mechanism of electroacupuncture treatment with Bushenyinao for AD. Method According to the "kidney and brain"theory,eight-month-old SAMR1 and SAMP8 mice were treated with electroacupuncture at "Baihui","Shenshu"and "Taixi"points for eight consecutive weeks. The paraffin section was made using the half of the mouse brain. The structure changes of Nissl body were observed by toluidine blue staining. Results The neurons were large and regular in the control group of SAMR1 mice. There were abundant Nissl bodies in the cytoplasm of neurons. In the model group of SAMP8 mice,the volume of neuronal decreased or lose the cell morphology. Nissl bodies were lighter in color and less in number. The volume of neurons become larger. Nissl body was dense and no obvious loss of vacuolar degeneration in acupuncture and medicine group. Conclusion Electroacupuncture with Bushenyinao based on the theory of"kidney and brain"can improve the morphological and functional status of neurons in SAMP8 mice,which may have a good therapeutic effect on AD.
