摘 要:目的利用低温空气等离子体反应装置,对模拟染菌载体聚氯乙烯(PVC)、聚四氟乙烯(PTFE)和聚对苯二甲酸乙二醇酯(PET)表面金黄色葡萄球菌的杀灭效果和机理进行研究。方法采用双悬浮探针对空气等子体的电子温度以及电子自旋共振波谱对自由基浓度进行了测定;采用扫描电子显微镜观察等离子体处理前后细菌细胞的形态变化。结果空气等离子体的灭菌效果符合放电区>余辉区>远程区的趋势;不同载体材料表面灭菌效果顺序与材料的亲水性一致;对数生长期的菌体比稳定期更容易被等离子体杀灭;有机污染物浓度增大,灭菌效果减弱;扫描电镜结果证实细菌细胞不同程度损伤,内容物逸出。结论等离子体灭菌过程是由各种活性粒子协同作用对细菌细胞造成破损的结果,而自由基在其中起主导作用,紫外光仅起辅助作用。Objective To study the germicidal efficiency of Staphylococcus aureus on the surface of medical poly( tetrafluoroethylene)( PTFE),Polyvinyl Chloride( PVC) and Polyethylene Terephthalate( PET) film with low temperature air plasma. Methods Double Langmuir probe and electron spin resonance spectroscopy were used to determine the temperature of electrons and the concentration of free radicals,respectively. The cell morphology was studied by scanning electron microscopy. Results The germicidal effects of low temperature air plasma,which was in according with the order of the surface wettability of the polymer material film,was discharge area post-glow area remote area. Cells of stationary growth phase were more sensitive than those of logarithm growth phase to the effects of plasma. With concentration of organic pollution augmenting,germicidal effect decreased. The result of SEM showed the crack of cell wall( cell membrane),the change of cell permeability and the leak of cell content. Conclusion The sterilization process of Staphylococcus aureus is the result of oxygen and all active factors working together,and free radicals play an important role in it,the action of UV radiation was slight.
