摘 要:目的:探讨补肾生髓方对衰老大鼠学习记忆能力及衰老相关蛋白P16、P21的影响。方法:将50只实验大鼠随机分成D-半乳糖衰老模型组、正常组、补肾生髓方组、六味地黄丸组、维生素E组共5个组。采用颈背部皮下注射D-半乳糖400mg/kg,连续4个月,制备衰老模型,同时给予相应药物灌胃,正常组给予等剂量0.9%氯化钠注射液。采用Morris水迷宫检测大鼠学习记忆能力,Western blot检测大鼠海马中P16、P21蛋白的表达。结果:补肾生髓方能够缩短D-半乳糖衰老大鼠到达平台的时间,降低衰老模型大鼠海马中P16、P21的含量。结论:补肾生髓方能够提高衰老大鼠的学习记忆能力,通过降低大鼠体内P16、P21的含量从而达到延缓衰老的目的。Objective:To investigate the effect of Bushen Shengsui prescription on learning and memory abilities and senescence - associated proteins P16 and P21 in a rat model of D - galactose - induced senescence. Methods:A total of 50 experimental rats were randomly divided into D - galactose - induced senescence model group, normal group, Bushen Shengsui prescription group, Liuwei Dihuang pill group, and vitamin E group. Subcutaneous injection of D - galactose at the back of the neck was performed for four consecutive months to establish a rat model of senescence and the corresponding drug was given by garage ;the rats in the normal group were given 0.9% sodium chloride injection at the same dose. The Morris water maze test was used to evaluate learning and memory abilities, and Western blot was used to measure the pro- tein expression of P16 and P21 in the hippocampus. Results:Bushen Shengsui prescription significantly shortened the time to reach the platform and reduced the content of P16 and P21 in the hippocampus in rats with D - galactose - induced se- nescence. Conclusion:Bushen Shengsui prescription can improve the learning and memory abilities of senescent rats and delay senescence by reducing the content of P16 and P21.
