摘 要:目的:观察六味地黄汤对肝微粒体色素P450酶系的影响,并对单味中药与其复方对色素P450酶系的影响进行比较分析。方法:ICR小鼠50只,雌雄各半,随机分成空白组(0.9%氯化钠注射液),肝药酶诱导剂组(苯巴比妥钠)、肝药酶抑制剂组(西米替丁)、六味地黄汤低剂量组[5.63g/(kg·d)]、六味地黄汤高剂量组[11.26g/(kg·d)],每组10只。苯巴比妥钠组为皮下注射给药,剂量为0.08g/(kg·d),其余组均为灌胃给药,西米替丁组剂量0.4g/(kg·d),其他中药组剂量均为20m L/(kg·d),1次/d。共10d。末次给药1h后处理动物,取肝组织制备肝微粒体,ELISA法测定细胞色素P450的含量,进行统计比较。结果:与空白组比较,六味地黄汤低、高剂量组色素P450含量均明显增加(P<0.05或P<0.01);高剂量组增加优于低剂量组(P<0.05)。肝药酶抑制剂组色素P450含量较空白组明显增加(P<0.01);肝药酶诱导剂组色素P450含量较空白组明显减少(P<0.05)。结论:六味地黄汤对肝微粒体色素有诱导作用,且高剂量组的作用强于低剂量组。Objective:To investigate the effect of Liuwei Dihuang decoction on liver microsomal cytochrome P450 through a comparative analysis of the effect of single versus compound Chinese medicine on P450. Methods : A total of 50 ICR mice(25 male and 25 female mice)were randomly divided into blank group(0. 9% sodium chloride injection) ,liver enzyme inducer group (phenobarbital sodium), liver enzyme inhibitor group (cimetidine), and low - and high -dose Li- uwei Dihuang decoction groups (5.63 and 11.26g,/kg · d, respectively) , with 10 mice in each group. Phenobarbital sodium was given by subcutaneous injection at a dose of 0. 08 g/kg · d ; the other drugs were given by gavage, with a dose of cimetidine of 0. 4g/kg · d and a dose of traditional Chinese medicine of 20mL/kg · d, once a day for 10 days. The animals were treated at 1 hour after the last dose, and liver tissue was collected to prepare liver microsome. ELISA was used to measure the content of cytochrome P450,and the results were analyzed and compared. Results:Compared with the blank group,the low - and high - dose Liuwei Dihuang decoction groups had a significant increase in cytochrome P450 ( P 〈 0. 05 or P 〈 0.01 ) , and the high - dose group had a significantly greater increase than the low - dose group( P 〈 0. 05 ). Compared with the blank group ,the liver enzyme inhibitor group had a significant increase in cytochrome P450 (P 〈 0. 01 )and the liver enzyme inducer group had a significant reduction in cytochrome P450 ( P 〈 0. 05 ). Conclusion : Liuwei Dihuang decoction has an inductive effect on liver microsomal cytochrome, and high - dose Liuwei Dihuang decoction has a stronger effect than low- dose Liuwei Dihuang decoction.
