摘  要:目的:研究草苁蓉多糖对四氯化碳所致HepG2细胞损伤的保护作用。方法:用四氯化碳诱导损伤HepG2细胞建立损伤模型,并以噻唑蓝法检测细胞存活率;以比色法检测细胞外液的乳酸脱氢酶(LDH)、谷丙转氨酶(ALT)和谷草转氨酶(AST)活性以及细胞还原型谷胱甘肽(GSH)、丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性和Caspase-3、Caspase-8、Caspase-9的相对活性。结果:与模型组比较,草苁蓉多糖组Hep G2细胞存活率升高;细胞外液中LDH、AST和ALT活性降低;细胞MDA含量降低,细胞GSH含量和SOD活性增高;细胞Caspase-3、Caspase-8和Caspase-9相对活性降低。结论:草苁蓉多糖对四氯化碳所致Hep G2细胞损伤具有保护作用。Objective: To investigate the protective effect of Boschniakia rossica polysaccharides on oxidative damage induced by carbon tetrachloride in HepG2 cells. Methods: HepG2 cells were exposed to carbon tetrachloride and used as the model of oxidative stress. And then the antagonist effect of Boschniakia rossica polysaccharides was detected by pretreatment with Boschniakia Rossica polysaccharides prior to carbon tetrachloride challenge. Cell viability was evaluated with MTT assay,and levels of LDH,ALT,AST,GSH,MDA,SOD and the relative activities of Caspase-3,Caspase-8 and Caspase-9 were measured by the microplate tecniques. Results: Carbon tetrachloride increased oxidative stress and decreased the cell viabilities of HepG2 cells. Compared with the model group,cell viabilities of Boschniakia rossica polysaccharide group increased in a dose-dependent manner.Boschniakia rossica polysaccharides also reduced the leakage of LDH,ALT and AST to culture fluid,reduced MDA content,increased GSH content and SOD activity,and reduced the relative activities of Caspase-3,Caspase-8 and Caspase-9. Conclusion: Boschniakia rossica polysaccharides have a protective effect on cellular oxidative damage induced by carbon tetrachloride in HepG2 cells.
