摘 要:目的:观察电针"曲池""足三里"穴对高血压前期Dahl盐敏感(DS)大鼠肾脏NO/c GMP/PKG信号通路的影响及组织形态的改变,探讨电针"曲池""足三里"对高血压前期肾脏损害保护作用机制。方法:盐抵抗大鼠(DR)10只作为正常组;DS大鼠30只,随机分为模型组、针刺组和非穴位组,每组10只。采用高盐饲料喂养的方法诱发高血压前期模型。电针组电针"曲池""足三里"穴,1次/d,6次/周,共5周。非穴位组取双侧髂嵴上10~15 mm、后正中线旁开20mm区段内一个固定对照点,治疗方法同电针组。测量大鼠治疗期间血压变化情况以及治疗后血清血清尿素氮(UREA)、血肌酐(CREA)、血尿酸(UA)含量;ELISA法检测NO含量;荧光定量PCR检测大鼠肾组织溶解型鸟苷酸环化酶(s GC)、蛋白激酶G(PKG)的mRNA表达;HE染色检测大鼠肾组织病理形态变化。结果:治疗期间,电针组大鼠血压逐步下降。与正常组相比,模型组血清UREA、CREA、UA含量显著升高(P<0.05),NO含量显著降低(P<0.05),s GC、PKG mRNA表达显著降低(P<0.05);与模型组相比,电针组血清UREA、CREA、UA含量显著降低(P<0.05),NO含量显著升高(P<0.05),s GC、PKG mRNA表达显著升高(P<0.05)。结论:电针能有效降低高血压前期DS大鼠血压,提高NO含量、c GMP、PKG基因表达,改善组织小管扩张、间质炎症细胞浸润等,这可能是电针通过NO/c GMP/PKG信号通路发挥对高血压前期DS大鼠肾脏保护作用机制之一。Objective: To observe electroacupuncture at Quchi( LI11) and Zusanli( ST36) of pre-hypertension in Dahl salt-sensitive rats( DS) kidney NO/c GMP/PKG signal pathway and the morphology change of electroacupuncture at Quchi and Zusanli of pre-hypertension renal damage protection mechanism. Methods: Ten Salt resistant rats( DR) were as the normal group and thirty DS rats were randomly divided into model group,acupuncture group and non-acupoint group,with 10 rats in each group. A high salt diet was used to induce the model of hypertension. The electroacupuncture group were given electroacupuncture at Quchi( LI11) and Zusanli( ST36),once a day,6 times/week,totally 5 weeks. Non-acupoint group took the area which was 10-15 mm up bilateral iliac crest and 20 mm away from the middle of the midline with the same electroacupuncture. Measure the changes in blood pressure( BP) after treatment,serum urea nitrogen( UREA),serum creatinine( CREA),blood uric acid( UA). ELISA method was used to detect the content of nitric oxide( NO) and fluorescence quantitative PCR to detect rat renal tissue soluble guanosine cyclic acid cyclase( s GC) and protein kinase G( PKG) mRNA expression. Hematoxylin and eosin( HE) staining was to detect the rat kidney tissue pathological change. Results: During the treatment,the blood pressure of the rats in the electroacupuncture group decreased gradually. Compared with the normal group,model group' s serum levels of UREA,CREA and UA increased significantly( P〈0. 05) and NO content decreased significantly( P〈0. 05). The s GC and PKG mRNA expression decreased significantly( P〈0. 05). Compared with the model group,electroacupuncture group's serum levels of UREA,CREA and UA decreased significantly( P〈0. 05) and NO levels increased significantly( P〈0. 05).
