小半夏汤对顺铂诱发大鼠脑干孤束核Fos蛋白表达的影响

摘　　要：目的观察小半夏汤对顺铂化疗大鼠脑干孤束核Fos蛋白表达的影响。方法雄性大鼠随机分为正常对照组(10ml·kg^(-1)蒸馏水i.g.)、顺铂模型组(0.1ml·kg^(-1)蒸馏水i.g.)、昂丹司琼治疗组(昂丹司琼2.6mg·kg^(-1),i.g.)、阿瑞吡坦治疗组(阿瑞吡坦11.1mg·kg^(-1),i.g.)、小半夏汤治疗组(小半夏汤1.6g·kg^(-1),i.g.),单笼饲养。模型组和各治疗组6 mg·kg^(-1)顺铂i.p.造模。从造模前3 d开始,各组动物每天按照上述剂量分别灌胃相应药物2次(间隔12 h)直至处死。注射顺铂后24 h、72 h深度麻醉大鼠,心脏灌注,取脑,冰冻切片,免疫组化技术观察各组大鼠孤束核内Fos阳性细胞数。结果与空白对照组比较,注射顺铂后24 h、72孤束核的Fos阳性细胞数均显著增加(P<0.001),造模后24h时间点的增加程度大于72 h。小半夏汤治疗组与顺铂模型组比较,两个时间点Fos阳性细胞数均显著降低(P<0.001);与空白对照组比较,小半夏汤治疗组Fos阳性细胞数有增加趋势,但无统计学意义(P>0.05);与两种阳性药组分别进行比较,小半夏汤治疗组Fos阳性细胞数没有显著性差异(P>0.05)。结论顺铂能够使大鼠孤束核Fos蛋白表达显著增加,小半夏汤可以抑制顺铂导致的孤束核Fos蛋白表达增加。Objective To elucidate whether cisplatin can cause Fos expression in the nucleus of the solitary tract（NTS） and the effect of XiaoBanXiaTang. Methods Immunohistochemistry method was used to investigate effect of different time after cisplatin treatment on Fos expression in NTS of rats, and the influence of XiaoBanXiaTang to the Fos expression in NTS neurons of rats in- duced by cisplatin treatment. Results Fos protein expression in NTS neurons significantly increased at 24h, 72h after cisplatin treatment. In addition, XiaoBanXiaTang antagonized the Fos protein expression in NTS neurons caused by cisplatin. Conclusion Cis- platin could significantly increase the expression of Fos protein in NTS neurons, which was time - dependent, and antagonized by XiaoBanXiaTang.
