摘  要:目的探讨枸杞多糖(Lycium barbarum polysaccharide,LBP)在力竭运动致大鼠胸主动脉血管内皮细胞损伤中的保护作用。方法雄性SD大鼠40只,按体重随机分为空白对照组,有氧运动组,力竭运动组,力竭运动+LBP组,进行5周游泳训练,建立力竭运动模型。采用离体血管环灌流技术测定大鼠胸主动脉血管环对去甲肾上腺素(NE)诱导的血管反应性;高倍镜下观察HE染色的大鼠胸主动脉血管组织形态的变化;流式细胞术检测外周循环血液中的内皮细胞数;激光共聚焦显微镜检测各组氧化应激(H_2O_2)后细胞内[Ca^(2+)]i浓度变化。结果灌服LBP可以抑制NE诱发的血管收缩反应(P<0.01),减少内皮细胞脱落,降低细胞内Ca^(2+)浓度。结论力竭运动会导致大鼠胸主动脉血管内皮细胞损伤脱落;LBP对大鼠胸主动脉内皮细胞损伤有一定的保护作用。Objective To discuss the protective effect of Lycium barbarum polysaccharide in the injury of vascular endothelial cells of thoracic aorta of exhaustive exercise in rats. Methods Forty male SD rats were randomly divided into the control group, aerobic exercise group, exhaustive exercise group, intervention group of Lycium barbarum polysaccharide. Each group was trained through swimming for five weeks. The response of rat aortic rings to NE were measured by isolated vascular ring perfusion tech- nique ; The changes of vascular organization form of rat thoracic aorta by HE staining under high power microscope ; Flow cytome- try was used to detect the number of endothelial cells in peripheral blood and laser confocal microscopy was used to test [ Ca2 ~ ] i concentration change in the cell cultures. Results LBP could inhibit the vasoconstriction induced by NE (P 〈 0.01 ), decrease the loss of endothelial cells, reduce the amplitude of intracellular Ca2+ concentration. Conclusion Exhaustive exercise could in- duce injury and damage loss in rat aortic endothelial cell; LBP has certain protective effect in rat aortic endothelial cell injury.
