滋肾青芪颗粒对胶原诱导性关节炎小鼠的抗炎作用研究

摘　　要：目的观察滋肾青芪颗粒对胶原诱导性关节炎小鼠的炎症预防及治疗作用。方法 DBA/1小鼠随机分为正常组,模型组,滋肾青芪颗粒低、中、高剂量组,甲氨蝶呤组。除正常组外,其余各组建立胶原诱导性关节炎小鼠模型,于攻击造模当天开始,连续给药3周。评定关节炎指数,观察小鼠关节病理切片炎性介质评分、骨破坏评分,ELISA法测定各组小鼠血清抗Ⅱ型胶原(CⅡ)特异性抗体水平。结果滋肾青芪各剂量组均可有效降低小鼠关节炎指数,与模型组比较差异有统计学意义(P<0.05);滋肾青芪高剂量组能有效降低小鼠血清抗CⅡ的抗体滴度,与模型组比较差异有统计学意义(P<0.05);滋肾青芪颗粒各组炎性介质评分、骨破坏评分均低于模型对照组(P<0.05)。结论滋肾青芪颗粒对胶原诱导性关节炎小鼠关节炎具有预防治疗作用,其机制可能与降低关节炎小鼠血清中特异性抗血清CⅡ抗体水平有关。Objective To observe the preventive and therapeutic effects of Zishen Qingqi（ ZSQQ） Granule on inflammation in collagen-induced arthritis mice. Methods DBA/1 mice were randomly divided into the normal group,model group,methotrexate group and ZSQQ groups with low-,middle-and high-dose. Except the normal group,the mice model of collagen-induced arthritis was established in the other groups. From the same day of induction,each group was treated with the corresponding drugs for continuous 3 weeks. The arthritis index was evaluated. The scores on inflammatory mediators and bone destruction in the pathological sections of mouse joint were observed.The serum level of anti-collagen type Ⅱ（ C Ⅱ） specific antibody was detected by ELISA. Results Compared with the model group,the arthritis index was effectively decreased in the ZSQQ groups with low-,middle-and high-dose,with statistically significant differences（ P〈0. 05）. Compared with the model group,the serum antibody titer of anti-CⅡ was effectively decreased in the ZSQQ group with high-dose,with statistically significant difference（ P〈0. 05）. The scores on inflammatory mediators and bone destruction in the ZSQQ groups with low-,middle-and high-dose were all lower than those in the model control group（ P〈0. 05）. Conclusion Zishen Qingqi Granule Shows the preventive and therapeutic effects in collagen-induced arthritis mice. Its mechanism maybe associated with the decrease in serum level of specific anti-CⅡ antibody in arthritis mice.
