摘  要:为探讨成纤维细胞生长因子21(FGF21)对布鲁菌感染的抗炎作用,本试验检测患布鲁菌病羊外周血中FGF21的含量;体外试验使用FGF21与布鲁菌共培养后检测细菌存活率;分析FGF21对Raw264.7细胞毒性,并以FGF21预处理Raw264.7细胞后,使用布鲁菌侵染细胞,观察细胞因子的释放以及TLR/MyD88信号通路的变化。结果显示,患布鲁菌病羊外周血中FGF21的含量显著升高,FGF21并不影响布鲁菌的存活率且对Raw264.7细胞没有毒性;FGF21预处理后可以抑制布鲁菌侵染诱导的IL-6、TNF-α、MCP1和LDH的释放,并且FGF21可提高IL-10的含量;同时FGF21可降低布鲁菌侵染引起的TLR/MyD88表达的升高。结果表明,患布鲁菌病羊外周血中FGF21的含量显著升高,FGF21通过TLR/MyD88通路缓解布鲁菌侵染诱导的炎性细胞因子的释放。To study the anti-inflammatory mechanism of fibroblast growth factor 21 (FGF21) in the process of Brucella infecting mice macrophage, the serum levels of FGF21 in brucellosis sheep were measured. The tested indicators included the efficiency of FGF21 on bactericidal analysis and cytotoxic effect on Raw264.7 cells,and the cytokines from Bruceila -infected Raw264.7 cells pretreated with FGF21. The results showed that brucellosis sheep increased circulating levels of FGF21. FGF21 did not affect the viability of Brucella and no cytotoxic effect on Raw264.7 cells. Pretreated FGF21 attenuated Brucella infection induced IL-6, TNF-α, MCP1 and LDH release, while FGF21 increased IL-10 release. Furthermore, FGF21 downregulated TLR/MyD88 signaling induced by the Brucella infection. The results indicated that Brucella infection elevated FGF21 levels in sheep. FGF21 exhibits inhibitory effect on the expression of pro-inflammatory cytokines via modulation of TLR/MyD88 pathway after Brucella infection.
