摘  要:目的观察KKAy糖尿病小鼠骨骼肌组织中叉头状因子(Fox)O1、过氧化物酶体增殖物激活受体(PPAR)γ的表达,探讨FoxO1的表达与糖脂代谢的关系。方法8周龄KKAy小鼠8只为糖尿病模型组(DM),予以高脂饲料喂养;8周龄C57BL/6J小鼠16只,随机分为空白对照组(NC)8只、高脂喂养组(HFD)8只。喂养至16 w后检测3组小鼠空腹血糖(FBG)、胰岛素(Ins)、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)及低密度脂蛋白胆固醇(LDL-C);采用实时荧光定量PCR(RT-PCR)技术检测骨骼肌组织FoxO1及PPARγmRNA表达,Western印迹检测骨骼肌组织中FoxO1蛋白表达。结果DM组FBG、TG、TC、LDL-C、Ins和HOMA-IR水平均较NC组及HFD组显著升高(P<0.05),HFD组小鼠Ins、TC、LDL-C和HOMA-IR较NC组显著升高(P<0.05);DM组PPARγmRNA较NC组及HFD组均显著降低(P<0.01),HFD组PPARγmRNA较NC组显著降低(P<0.05);DM组FoxO1 mRNA及蛋白表达较NC组及HFD组均明显增高(P<0.05)。DM组骨骼肌FoxO1 mRNA表达与FBG(r=0.910,P<0.01)、Ins(r=0.864,P<0.01)、TG(r=0.897,P<0.01)和HOMA-IR(r=0.944,P<0.01)呈正相关,与PPARγmRNA呈负相关(r=-0.719,P<0.01)。结论FoxO1的表达与糖血脂代谢密切相关。Objective To observe the expression of FoxO1 and PPARγ inskeletal muscle of KKAy mice and explore the relation- ship between FoxO1 and glycolipid metabolism. Methods KKAy mice with the high-fat diet were chosen into the diabetic model group (DM) (n= 8) ,C57BL/6J mice were randomly divided into control group (NC) (n = 8 )and the high-fat diet group (HFD) (n = 8). The blood samples were used for measuring fasting blood glucose (FBG), serum insulin (Ins), triglyceride (TG), total cholesterol (TC), high density lipoprotein-C(HDL-C) and low density lipoprotein-C (LDL-C). 16 weeks later, insulin resistance was assessed by HOMA-IR. The FoxO1 and PPARγ/mRNA expression levels in skeletal muscle were detected by real time quantitative(RT-PCR). The FoxO1 protein expression was detected by Western bolt method. Results Levels of FBG, TG, TC, LDL-C, Ins and HOMA-IR were significantly higher in DM group than those in control group and high-fat diet group. Levels of Ins, TC, LDL-C and HOMA-IR were significantly higher in the high-fat diet group than those in control group. The mRNA expression of PPARγ in skeletal muscle of DM group(0.56±0.05) was reduced when compared with control group (1.90±0.13 )and the high-fat diet group (0.91 ±0.06) (P〈0.01 ), the PPARγ mRNA expression in the high-fat group was lower than that of the control group( P〈0. 05 ). The mRNA level of FoxO1 in skeletal muscle of DM group(48.47±2.46)was higher than that of control group( 18.26±2.49 )and the high-fat diet group(20.00±2.06)( P〈0.05 ). The protein expression of FoxO1 in skeletal muscle of DM group (4.73 ± 079 ) was higher than that of control group (0..91 ±0. 17 )and the high-fat diet group (1.26 ±0. 16 ) (P〈0.05). Moreover, the mRNA expression of FoxO1 in skeletal muscle was positively correlated with FBG( r = 0. 910, P〈0. 01 ), Ins( r = 0. 864, P〈0.01 ), TG ( r = 0. 897, P〈0. 01 ) and HOMA-IR ( r = 0. 944,
