摘  要:目的研究通过建立石英致小鼠肺部急性炎性反应模型,探讨外源性加入CC16蛋白对石英致小鼠肺部急性炎性反应的作用及可能的机制。方法将64只小鼠随机分为4组,石英粉尘悬液气管滴注染尘;4组小鼠分别为生理盐水、石英组、低剂量CC16补充组和高剂量CC16补充组;于染尘后1 d、7 d处死,收集肺组织及肺部灌洗液;检测肺部灌洗液中IL-1β、CC16的表达水平及肺组织中Caspase-1蛋白表达水平;H&E染色后观察肺组织炎性程度。结果染尘3组(石英组、低剂量CC16补充组,高剂量CC16补充组)小鼠肺部灌洗中的CC16表达水平较生理盐水组显著降低,且差异具有统计学意义(P<0.05);染尘后7d,与生理盐水组相比,染尘3组小鼠肺组织中Caspase-1蛋白表达水平明显升高,且高剂量CC16蛋白补充组与石英染尘组小鼠相比,其肺组织中Caspase-1蛋白表达水平显著降低(P<0.05);染尘后1d,与石英组小鼠相比,CC16蛋白补充组小鼠IL-1β表达水平明显降低,随着CC16补充剂量增加,IL-1β表达水平明显降低,存在剂量-反应关系。结论 CC16可能在石英致小鼠肺组织急性炎性反应中起到一定作用;CC16可能通过NAPL3相关通路,抑制caspase-1表达水平,降低前炎性因子IL-1β的表达水平,从而减轻炎性反应。Objective To assess the expression and potential mechanism of CC16 in silica-induced acute inflammatory response in mice. Methods A total of 64 C57BL/C mice were randomly divided into 4 groups, inc1uding nor^nal saline group, si1ica group, silica + low - dose CC16 protein group,and silica + high - dose CC16 protein group. On the 1st day, 7th day after instilled treatment, 16 mice of each group were sacrificed. The CC16 and IL - 1β concentrations in bronchoalveolar lavage fluid(BALF) were measured by enzyme linked immunosorbent assay(ELISA);RT - PCR w,as performed to detect the relative expressions of IL -1β and Caspase - 1 mRNA in lung tissues;Westem - Blot was used to detect the expressions of Caspase - 1 protein in lung tissues; H&E staining was observed to investigate the pathological changes and inflammatory response in mice. Results Compared with the normal saline group,the silica-treated groups(silica group,silica + low -dose CC16 protein group and silica + high -dose CC16 protein group) had a lower concentration of CC16 in BALF( P 〈 0 . 0 5 ) ;while on the 7th day after trea tmen t, the relative expressions of Caspase - 1 mRNA increased among the silica-treated groups ( P 〈 0. 0 5 ) . Compared with silica group,the high-dose CC16 protein group had a lower expression of Caspase - 1 mRNA in lung tissues. Compared with saline group, the concentration and the relative mRNA expression of IL -1βincreased among the silica-treated groups,and compared with the silica group,the relative expressions of IL - 1β mRNA decreased with increasing CC16 concentrations among the silica-treated groups. Conclusions CC16 played a potential role in silica- induced acute inflammatory^ response; CC16 may decrease Caspase - 1 expressions and IL - 1 β concentration,through NAPL3 signal pathway,thereby alleviate the acute inflammation response.
