摘 要:目的探讨知柏地黄汤对大鼠血清及尿液差异代谢物的影响。方法将正常对照组和知柏地黄汤组大鼠分别予生理盐水和知柏地黄汤水煎剂进行灌胃21d后,取尿液和血清处理后,采用EclipsePlusC18色谱柱(2.1×50mm,1.8μm);柱温为35℃;流动相A:0.1%甲酸水溶液,流动相B:0.1%甲酸乙腈溶液,流速:0.2ml/min。结果在血清样本中包括脂肪酸生物合成,甘油磷脂代谢和亚油酸代谢途径在内的三种代谢途径发生明显变化;尿液样品中的缬氨酸,亮氨酸和异亮氨酸生物合成途径,甘油磷脂代谢和苯丙氨酸,酪氨酸和色氨酸生物合成均有显著变化。结论知柏地黄汤组大鼠尿液中缬氨酸和亮氨酸的增加可进一步证实知柏地黄汤能提供氧化能量,促进蛋白质合成,提高免疫功能。Objective To investigate the effect of Zhibai Dihuang Decoction on differences of serum and urine metabolites in rats. Methods The normal control group and Zhibai Dihuang Decoction group were treated with normal saline and Zhibai Dihuang decoction for 21 days. After taking urine and serum, the samples were treated with Eclipse Plus C 18 ( 2.1 ×50mm,1.8μ m ) .Column temperature was 35℃. Mobile phase A: 0.1% formic acid aqueous solution, mobile phase B: 0.1% formic acid acetonitrile solution at a flow rate of 0.2ml/min. Results Three metabolic pathways in the serum samples including fatty acid biosynthesis, glycerophospholipid metabolism and hnoleic acid metabolism pathway were significantly altered. The valine, leucine and isoleucine biosynthesis in urine samples and glycerol phospholipid metabolism and phenylalanine, tyrosine and tryptophan biosynthesis pathway were significantly changed. Conclusion The increase of valine and leucine in urine of Cynanchum may further confirm whether Zhibai Dihuang Decoction can provide oxidative energy, promote protein synthesis and improve immune function.
