摘 要:目的利用超高效液相色谱-四级杆-飞行时间串联质谱(UHPLC-Q-TOF MS)对精神分裂症患者和健康对照人群的尿液进行代谢组学研究,寻找精神分裂症患者尿液中的差异代谢物,为精神分裂症患者代谢综合征的早期筛查提供科学依据。方法尿液样品经稀释过滤后进样分析,利用主成分分析方法分析所采集的数据。结果根据谱库共筛选了7个表征标记物,并进行了结构鉴定。结果发现,精神分裂症患者较对照组尿液代谢表型有显著差异,主成分分析得分散点图中对照组与精神分裂症患者组分类良好。结论精神分裂症患者糖、脂、氨基酸等代谢通路均发生紊乱,可能在代谢综合征发生发展中有重要作用。Objective An ultra-performance liquid chromatography quadrupole time of flight mass spectrometric (UHPLC-Q-TOF MS) method is developed for the study on metabolic variations of urine from schizophrenia patients and healthy volunteers.And the study will provide a scientific basic for early screening of schizophrenia comorbied with metabolic syndrome. Methods The urine samples would analyze after they were diluted and filtered, then principal component analysis method was used on the collected data.Results A total of 7 characterization markers were selected and the structural identification was carried out. PLS-DA was used for group differentiation and potential biomarkers selection. As shown in the scores plot, the distinct clustering among schizophrenia patients and healthy volunteers was observed. Conclusion It is suggested that the metabolic pathways such as carbohydrate, lipid and amino acid in schizophrenia can be disturbed, and show the metabolic disturbance in schizophrenia patients may play an important role in the development of metabolic syndrome.
