摘  要:目的建立重要的红细胞血型的基因分型技术用于筛选抗原谱合理的试剂红细胞。方法首先建立检测红细胞血型系统Rh CE、Kidd、MNS、Duffy、Diego、Lutheran、Colton、Lutheran和Kell等的聚合酶链式反应-序列特异性引物(PCR-SSP)分型技术;对于无法获得相关血型试剂的Diego和Colton血型系统,采用测序方法验证该技术检测血型的准确性,其他6个血型系统则用血清学方法验证该技术检测血型的准确性;最后应用该方法对82名O型固定献血者的8个血型系统做基因分型。结果血型基因PCR-SSP分型技术的结果,与血清学和测序检测结果比较,完全一致。所建立的血型基因分型技术对82名O型献血者8种红细胞血型检测,筛选出13位可能适合提供试剂红细胞的献血者。结论成功建立了分型准确、方便、经济的红细胞血型基因PCR-SSP技术,用于从大量献血者中筛选出抗原谱合理的试剂红细胞。Objective To establish genotyping methods for important blood groups and screen antigens on reagent red cells. Methods Polymerase chain reaction-sequence specific primers( PCR-SSP) genotyping methods were established for initial analysis important red blood systems( RHCE,Kidd,MNS,Duffy,Diego,Lutheran,Colton,Kell blood systems). As for Diego and Colton blood systems,genotyping was achieved by direct sequencing methods due to unavailability of corresponding reagent antibody. In the other 6 blood systems,Genotyping was validated using serologic typing. 82 immobilized O blood donors were genotyped for the eight blood systems by the two methods mentioned prior. Results The genotyping results were consistent with serological and sequencing results. Samples from 13 out of the 82 immobilized O blood donors were considered reagent-ready red cells. Conclusion This study has successfully established an accurate,convenient and low-cost PCR-SSP genotyping method,which can screen out reagent red cells from a large number of blood donors with a sensible antigen panel design.
